【摘要】：目的通過去細胞技術(shù),將帶一部分膀胱壁的大鼠輸尿管去細胞后,移植于另一只鼠膀胱壁上,在不重新種植細胞的情況下,利用機體作為"自體反應(yīng)器"來重構(gòu)支架,最終使輸尿管再生。方法20只SD大鼠的輸尿管經(jīng)過Triton-x100和SDS去細胞后用去離子水漂洗并經(jīng)過殺菌,支架經(jīng)過形態(tài)學(xué)鑒定后移植入其他鼠中,行膀胱修補術(shù)同時遠端結(jié)扎,經(jīng)過1、2、3、4周獲取移植物進行HE染色,免疫熒光法觀察分析。結(jié)果移植輸尿管被分成3個部分:粘附于膀胱的部分(B)、中間部分(M)、游離端(D)。發(fā)現(xiàn)4周后B部分有上皮細胞和平滑肌細胞,M部分有脂肪細胞,D部分被纖維化并且在4周后仍然無細胞。B部分在1~2周后,上皮細胞出現(xiàn),4周后,平滑肌細胞出現(xiàn)。結(jié)論未播種輸尿管在接合區(qū)域也能實現(xiàn)再生,并且先出現(xiàn)上皮層后出現(xiàn)肌層。
[Abstract]:Objective to transplant rat ureteral cells with a part of bladder wall into another rat bladder wall by means of acellular technique, and to reconstruct the scaffold by using the organism as an "autoreactor" without reimplantation of cells. Methods the ureter of 20 Sprague-Dawley rats was treated with deionized water after the removal of cells by Triton-x100 and SDS. The stents were implanted into other rats after morphological identification. Bladder repair was performed and distal ligation was performed. The grafts were harvested for HE staining after 4 weeks of operation. Results the ureter grafts were divided into three parts: the part of the ureter adhered to the bladder, the middle part of the ureter, and the free end of the ureter. It was found that there were epithelial cells in part B and adipocytes in part M of smooth muscle cells 4 weeks later. The D part is fibrotic and is still cell-free 4 weeks later. Part B is 1 to 2 weeks later. Conclusion the unseeded ureter can also regenerate in the junction region and the muscle layer after the appearance of the upper cortex.
