制備組織工程脫細胞真皮修復皮膚潰瘍創(chuàng)面的愈合機制

【摘要】:背景:近年來,人尸體脫細胞真皮在真皮缺損、軟組織充填中得到應用,但是該修復材料價格昂貴,來源相對有限。目的:分析組織工程脫細胞真皮修復皮膚潰瘍創(chuàng)面的效果。方法:隨機對照方法將S D大鼠分為自體未脫細胞網(wǎng)狀層真皮組和脫細胞真皮組。采用低滲溶液、2.5 g/L胰蛋白酶、體積分數(shù)0.5%Triton X-100和PBS方法制備組織工程脫細胞真皮,建立大鼠皮膚潰瘍創(chuàng)面模型,自體未脫細胞網(wǎng)狀層真皮組移植自體斷層皮片進行覆蓋,脫細胞真皮組采用新型顆粒狀組織工程脫細胞真皮進行修復,比較兩組修復效果及對皮膚潰瘍創(chuàng)面愈合的機制。結果和結論:(1)獲得的組織工程脫細胞真皮為白色狀,常溫下大體觀察呈顆粒狀;(2)蘇木精-伊紅染色顯示組織工程脫細胞真皮內部呈現(xiàn)出"樹枝狀"分布,并且膠原纖維束的間隙相對較大;(3)脫細胞真皮組植皮后2,3,4,8周創(chuàng)面移植皮膚成活率,顯著高于自體未脫細胞網(wǎng)狀層真皮組(P0.05);(4)脫細胞真皮組大鼠植皮后3,4,8周創(chuàng)面收縮率顯著高于自體未脫細胞網(wǎng)狀層真皮組(P0.05);(5)脫細胞真皮組創(chuàng)周正常皮膚膠原纖維束平均直徑及間隙率顯著高于自體未脫細胞網(wǎng)狀層真皮組(P0.05)。(6)結果提示:組織工程脫細胞真皮制備方法相對簡單,制備條件溫和,且制備的組織工程脫細胞真皮形態(tài)規(guī)則,修復皮膚潰瘍創(chuàng)面效果理想,能降低真皮內D NA殘留量,能顯著改善體內某些生物力學特性,促進創(chuàng)面愈合。
[Abstract]:Background: in recent years, human cadaveric acellular dermis has been used in dermis defect and soft tissue filling, but the material is expensive. Objective: to analyze the effect of tissue engineering acellular dermis on the repair of skin ulcer wound. Methods: SD rats were randomly divided into two groups: autologous unacellular reticular layer dermis group and acellular dermis group. With a hypotonic solution of 2.5 g / L trypsin, Tissue engineering acellular dermis was prepared by Triton X-100 and PBS method. The rat model of skin ulcer was established. Acellular dermis group was repaired with new granular tissue engineering acellular dermis. The effect of the two groups and the mechanism of wound healing of skin ulcer were compared. Results and conclusion the tissue engineered acellular dermis was white. At room temperature, the results of hematoxylin and eosin staining showed that there was a "dendritic" distribution in the acellular dermis of tissue engineering. In acellular dermis group, the survival rate of skin grafts was 8 weeks after skin grafting. The rate of wound contraction in acellular dermis group was significantly higher than that in unacellular reticular layer group (P0.05). The contraction rate of wound in acellular dermis group was significantly higher than that in non-acellular reticular layer group at 8 weeks after skin grafting. The average diameter and interspace rate were significantly higher than those in the unacellular reticular layer group (P0.05. 6). The results showed that the preparation method of tissue engineering acellular dermis was relatively simple. The condition of preparation is mild, and the prepared tissue engineering acellular dermis is regular, the effect of repairing skin ulcer wound is ideal, the content of DNA in dermis can be reduced, some biomechanical properties in vivo can be improved and wound healing can be promoted.
