【摘要】:為了優(yōu)化替代眼刺激動物試驗的牛角膜渾濁和滲透性(BCOP)實驗的方法,提高其適用范圍和預(yù)測性,選取新鮮牛眼角膜,直接暴露液體化學(xué)受試物作用10 min,后孵育2 h并用熒光素鈉處理90 min,采用濁度儀檢測角膜渾濁度,用酶標(biāo)儀檢測熒光素鈉滲透性,同時將角膜常規(guī)固定和制備組織學(xué)切片,觀察組織學(xué)病理變化并評分,基于上述3個指標(biāo)建立預(yù)測模型。結(jié)果顯示,將角膜渾濁度和滲透性2個定量指標(biāo),與角膜上皮組織、基質(zhì)組織和內(nèi)皮細(xì)胞的組織學(xué)變化評分進(jìn)行組合,可對輕微刺激到中等刺激的范圍進(jìn)行準(zhǔn)確細(xì)分。研究表明,結(jié)合組織學(xué)觀察的優(yōu)化BCOP方法可提高其預(yù)測性,適用于眼刺激機(jī)制研究和化妝品評估。
[Abstract]:In order to optimize the eye irritation test instead of animal bovine corneal opacity and permeability (BCOP) experiment method, improve the accuracy of its scope and selection of fresh bovine cornea, directly exposed to liquid chemical test for 10 min after 2 h incubation with fluorescein sodium treatment for 90 min, the turbidity meter to detect corneal opacity standard test, fluorescein sodium permeability by enzyme, while the conventional preparation and corneal histological observation, histological changes and pathological score, the above 3 indicators based on prediction model is established. The results showed that the corneal turbidity and permeability of 2 quantitative indicators, and the corneal epithelium, stroma endothelial cells and histological changes were combined, can accurately segment scope of slight irritation to moderate stimulation. The results show that the optimized BCOP method combined with histological observation can improve the predictive, suitable for eye stimulation machine System research and cosmetic evaluation.
