泡球蚴體外培養(yǎng)模型的建立及細(xì)胞親和性觀察

【摘要】: 目的:利用肝癌細(xì)胞(Bel-7404)、大腸癌細(xì)胞(Caco-2)和宮頸癌細(xì)胞(Hela)及其培養(yǎng)上述三種細(xì)胞的上清液,在體外對(duì)泡球蚴進(jìn)行培養(yǎng),建立穩(wěn)定的體外培養(yǎng)模型,觀察泡球蚴的在體外的細(xì)胞親和性及生長(zhǎng)、發(fā)育過程。方法:培養(yǎng)肝癌細(xì)胞(Bel-7404)、大腸癌細(xì)胞(Caco-2)和宮頸癌細(xì)胞(Hela),留取細(xì)胞上清液。取泡球蚴組織,分別與上述三種細(xì)胞及含三種細(xì)胞培養(yǎng)上清的DMEM完全培養(yǎng)基于37℃5%CO2培養(yǎng)箱中培養(yǎng)38d。實(shí)驗(yàn)期間對(duì)囊泡進(jìn)行計(jì)數(shù),觀察囊泡生長(zhǎng)情況,記錄囊泡最大、最小直徑,繪制生長(zhǎng)曲線。培養(yǎng)結(jié)束后,取培養(yǎng)囊泡囊液進(jìn)行蛋白定量。利用SDS-PAGE技術(shù)觀察囊液中蛋白條帶特征。結(jié)果:泡球蚴與三種細(xì)胞及其上清液中均能共同生長(zhǎng),呈出芽生殖方式,囊泡直徑0.5~5.0 mm;囊液蛋白含量分別為:肝癌細(xì)胞,3.8mg/ml;肝癌細(xì)胞上清液,1.1mg/ml;大腸癌細(xì)胞,3.2mg/ml;大腸癌細(xì)胞上清液,1.2mg/ml;宮頸癌細(xì)胞,2.5mg/ml;宮頸癌細(xì)胞上清液,3.8mg/ml。泡球蚴在體外與不同的細(xì)胞共培養(yǎng)的初期生長(zhǎng)特點(diǎn)基本相似:均在培養(yǎng)初期呈較快速性生長(zhǎng)(1~15d);生長(zhǎng)中期可見一平臺(tái)期(15~26d);但后期肝癌細(xì)胞及其上清液內(nèi)培養(yǎng)泡球蚴囊泡保持一個(gè)較穩(wěn)定囊泡數(shù)量,而其他結(jié)果則表現(xiàn)為逐漸減少。并對(duì)肝癌細(xì)胞上清液所培養(yǎng)的泡球蚴囊壁DNA經(jīng)PCR擴(kuò)增出Em特異性200bp條帶。SDS-PAGE觀察體內(nèi)外培養(yǎng)囊液中蛋白在14、47-66kd處有相似條帶。結(jié)論:1)泡球蚴體外生長(zhǎng)因素可不依賴培養(yǎng)細(xì)胞本身;2)由于體外與動(dòng)物體內(nèi)的培養(yǎng)環(huán)境的不同,及不同的共培養(yǎng)細(xì)胞可能引起囊泡內(nèi)囊液的蛋白含量有所差異;3)泡球蚴在體外與肝癌細(xì)胞及其上清液的親和性與其它二種細(xì)胞略有不同;4)體內(nèi)、外培養(yǎng)的囊泡含有相似蛋白;5)利用肝癌細(xì)胞、大腸癌細(xì)胞和宮頸癌細(xì)胞及其上清液建立泡球蚴體外培養(yǎng)模型具有一定的可行性,但可能存在一定的差異性。
