樹突狀細胞共刺激分子表達與免疫抑制劑的干預

【摘要】:背景:以往關(guān)于器官移植免疫抑制和抗排斥反應的研究,多關(guān)注T淋巴細胞介導的免疫反應及免疫抑制劑對T淋巴細胞的作用,樹突狀細胞作用尚不清楚,且對于免疫抑制劑對樹突狀細胞影響的表現(xiàn)及機制亦不盡相同。目的:比較不同免疫抑制劑對樹突狀細胞共刺激分子表達及功能的影響,探討其免疫抑制作用機制。方法:在誘導C57BL/6小鼠骨髓細胞定向分化為樹突狀細胞時分別加入20μg/L雷帕霉素、0.04 mg/L霉酚酸酯、10μg/L他克莫司和1 mg/L環(huán)孢霉素A。結(jié)果與結(jié)論:流式細胞儀檢測結(jié)果顯示,各組CD40表達為:雷帕霉素他克莫司環(huán)孢霉素A霉酚酸酯(P0.01);CD86表達分別為:雷帕霉素他克莫司環(huán)孢霉素A=霉酚酸酯(P0.01);CD80、CD11c及主要組織相容性復合體Ⅱ的表達各組差異無顯著性意義(P0.05)。單向混合淋巴細胞反應結(jié)果顯示,各組樹突狀細胞共刺激T細胞增殖能力表現(xiàn)為:雷帕霉素霉酚酸酯他克莫司=環(huán)孢霉素A(P0.05)。結(jié)果證實,雷帕霉素、他克莫司、環(huán)孢霉素A、霉酚酸酯均通過抑制樹突狀細胞共刺激分子CD40及CD86表達而發(fā)揮免疫抑制作用,以雷帕霉素最為顯著;雖然以上藥物對樹突狀細胞抗原遞呈能力主要組織相容性復合體Ⅱ的表達均無明顯抑制性,但均能抑制樹突狀細胞刺激T細胞增殖的能力,雷帕霉素的抑增殖能力最強。
