【摘要】：目的:在原核系統(tǒng)內(nèi)獲得高表達的人血小板衍生生長因子BB(PGDF-BB),并對形成的包涵體進行復性。方法:對PGDF-BB核酸編碼序列進行優(yōu)化,構(gòu)建pET-22b-PGDF-BB表達載體,以提高PCDF-BB的表達量;優(yōu)化PGDF-BB包涵體復性條件,提高蛋白復性率和生物活性。結(jié)果:構(gòu)建了pET-22b-PGDF-BB高效表達載體,原核表達的重組人PGDF-BB占細菌總蛋白的25%,PGDF-BB包涵體復性率達到15%。結(jié)論:對表達序列的優(yōu)化設計可顯著提高蛋白的表達量,復性方法的改良提高了蛋白的復性率和生物活性。
[Abstract]:Aim: to obtain highly expressed human platelet-derived growth factor BB (PGDF-BB) in prokaryotic system and renaturation of the inclusion bodies. Methods: the PGDF-BB nucleic acid coding sequence was optimized to construct the pET-22b-PGDF-BB expression vector to increase the expression of PCDF-BB, to optimize the PGDF-BB inclusion body renaturation conditions and to improve the protein renaturation rate and biological activity. Results: the high efficient expression vector of pET-22b-PGDF-BB was constructed. The recombinant human PGDF-BB expressed in prokaryotic expression accounted for 25% of the total bacterial protein. The inclusion body renaturation rate of PGDF-BB reached 15%. Conclusion: the optimized design of expression sequence can significantly increase the expression of protein, and the improvement of renaturation method can improve the renaturation rate and biological activity of protein.
