【摘要】： 目的:探討人脂肪源干細(xì)胞( adipose derived stem cells,ADSCs)在體外分離、培養(yǎng)、擴(kuò)增的方法及擴(kuò)增后向成骨細(xì)胞誘導(dǎo)分化與免疫調(diào)節(jié)的能力。方法:以組織消化的方法分離脂肪組織中的干細(xì)胞體外擴(kuò)增培養(yǎng),擴(kuò)增后的ADSCs在成骨誘導(dǎo)條件下促進(jìn)其向成骨細(xì)胞的分化,以明確其分化能力。同時檢測擴(kuò)增后的ADSCs在體外抑制淋巴細(xì)胞增殖的能力,以評價ADSCs的免疫原性及免疫調(diào)節(jié)功能。結(jié)果:體外分離培養(yǎng)的脂肪源干細(xì)胞生長穩(wěn)定,擴(kuò)增速度快。流式細(xì)胞儀檢測后顯示高表達(dá)干細(xì)胞相關(guān)抗原。向成骨細(xì)胞誘導(dǎo)后經(jīng)免疫組化染色及RT-PCR分析顯示誘導(dǎo)的細(xì)胞分泌成骨細(xì)胞的特異性細(xì)胞外基質(zhì)并形成鈣結(jié)節(jié)。同時擴(kuò)增后的ADSCs在體外不會引起淋巴細(xì)胞的增殖并且在混合淋巴細(xì)胞反應(yīng)中可以抑制淋巴細(xì)胞的增殖。結(jié)論:脂肪源干細(xì)胞在體外分離培養(yǎng)簡單,擴(kuò)增速度快,并且擴(kuò)增后仍具有定向成骨分化及免疫調(diào)節(jié)的能力,是可靠的組織修復(fù)和細(xì)胞治療的種子細(xì)胞來源。
[Abstract]:Aim: to investigate the methods of isolation, culture and amplification of human adipose stem cells (adipose derived stem cells,ADSCs) in vitro and their ability to induce differentiation and immunomodulation into osteoblasts. Methods: stem cells from adipose tissue were isolated by tissue digestion and cultured in vitro. The expanded ADSCs promoted the differentiation of adipose cells into osteoblasts under the condition of osteogenesis. In order to evaluate the immunogenicity and immunomodulatory function of ADSCs, the ability of expanded ADSCs to inhibit lymphocyte proliferation in vitro was also detected. Results: the adipose stem cells isolated and cultured in vitro grew stably and expanded quickly. High expression of stem cell associated antigen was detected by flow cytometry. After induction to osteoblasts, immunohistochemical staining and RT-PCR analysis showed that the induced cells secreted the specific extracellular matrix of osteoblasts and formed calcium nodules. Simultaneously amplified ADSCs did not induce lymphocyte proliferation in vitro and inhibited lymphocyte proliferation in mixed lymphocyte reaction. Conclusion: adipose-derived stem cells are easy to be isolated and cultured in vitro and have the ability of directional osteogenesis differentiation and immunomodulation, which is a reliable source of seed cells for tissue repair and cell therapy.
