【摘要】：目的:尿道括約肌系統(tǒng)是哺乳類動(dòng)物實(shí)現(xiàn)控尿的基本解剖結(jié)構(gòu),其中環(huán)繞尿道的橫紋括約肌是構(gòu)成尿道括約肌系統(tǒng)的最重要結(jié)構(gòu)成分。尿道括約肌系統(tǒng)的控尿功能表現(xiàn)得復(fù)雜而精細(xì),以滿足動(dòng)物在復(fù)雜多樣的日�；顒�(dòng)中對(duì)控尿的需求。研究尿道橫紋括約肌纖維的類型和相對(duì)含量,可以更好地理解尿道橫紋括約肌發(fā)揮復(fù)雜而精細(xì)的控尿功能的生理機(jī)制。 方法:取正常成年未生育雌性Fisher 344大鼠12只,年齡12月,麻醉后解剖取出完整尿道,提取尿道總RNA,采用逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)法檢測(cè)尿道橫紋括約肌肌球蛋白重鏈同源分子mRNA的類型和相對(duì)含量,從而獲得尿道橫紋括約肌纖維類型及其相對(duì)含量的資料。 結(jié)果:正常成年未生育雌性大鼠尿道橫紋括約肌表達(dá)四種肌球蛋白重鏈同源分子mRNA,即MHC-Ⅰ、MHC-Ⅱa、MHC-Ⅱb和MHC-Ⅱx。其中MHC-Ⅱb的mRNA相對(duì)含量最高(38.1±0.7)%,MHC-Ⅱa次之(34.6±0.8)%,MHC-Ⅱx再次之(18.1±1.2)%,MHC-Ⅰ最低(9.2±0.4)%。 結(jié)論:正常成年未生育雌性大鼠尿道橫紋括約肌表達(dá)全部4種肌球蛋白重鏈同源分子,顯示尿道橫紋括約肌含有4種類型的纖維:typeⅠ(慢收縮纖維)、typeⅡa(快收縮抗疲勞纖維)、typeⅡb(快收縮易疲勞纖維)和typeⅡx(超快收縮纖維)。尿道橫紋括約肌多樣化的纖維類型是其實(shí)現(xiàn)復(fù)雜和精細(xì)控尿功能的分子基礎(chǔ)。
[Abstract]:Objective: the urethral sphincter system is the basic anatomical structure of the mammalian urinary control, and the transverse sphincter around the urethra is the most important structural component of the urethral sphincter system. The urinary control function of urethral sphincter system is complex and delicate to meet the needs of animals in complex and diverse daily activities. To study the types and relative contents of sphincter fibers in urethra can better understand the physiological mechanism of the complex and elaborate urinary control function of the sphincter of urethra. Methods: 12 normal adult infertile female Fisher 344 rats, aged 12 months, were dissected after anaesthesia, and the complete urethra was dissected.
