【摘要】：為了解山東省2011年分離的1株脊髓灰質(zhì)炎(脊灰)疫苗高變異株的基因特征,并對該病例接觸者標本進行病原學鑒定及分析。本研究對采集的2份病例及40份接觸者糞便標本進行病毒分離,對脊灰病毒進行全基因組序列測定和分析,對非脊灰腸道病毒進行VP1區(qū)序列測定,并將分離率較高的柯薩奇病毒(Coxsackievirus,CV)B1和CV-B3與國內(nèi)外其他地區(qū)分離株進行VP1區(qū)同源性及系統(tǒng)進化樹分析。其中病例標本中分離到1株脊灰病毒(P1/11186),1株CV-A4和1株CV-A8;接觸者標本中分離到1株CV-A2、10株CV-B1、5株CV-B3和埃可病毒(Echovirus,E)3型、12型、14型各1株,提示當?shù)乜赡艽嬖谀c道病毒流行,應加強監(jiān)測。P1/11186為Ⅰ型脊灰疫苗病毒,在2A及3A區(qū)分別與Ⅱ型和Ⅲ型脊灰病毒發(fā)生重組,與相應的疫苗株相比全基因組共有25個核苷酸、9個氨基酸發(fā)生變異,其中VP1區(qū)8個核苷酸、5個氨基酸發(fā)生突變。CV-B1和CV-B3的同源性分析顯示10株CVB1之間核苷酸同源性為97.0%~100%,氨基酸同源性為98.9%~100%;5株CV-B3之間核苷酸與氨基酸同源性分別為92.6%~100%與99.2%~100%。CV-B1與CV-B3VP1完整編碼區(qū)進化樹分析顯示,山東分離株與國內(nèi)其他省份分離株親緣關系較近,位于同一基因簇,與世界其他地區(qū)分離株位于不同分支。
[Abstract]:In order to understand the gene characteristics of a poliomyelitis (polio) vaccine high variant strain isolated in Shandong Province in 2011, pathogenic identification and analysis were carried out on the samples of this case. In this study, the virus was isolated from 2 cases and 40 feces from contacts, the whole genome sequence of poliovirus was determined and analyzed, and the VP1 region sequence of non-polio enterovirus was sequenced. Coxsackievirus CV B1 and CV-B3 with high isolation rate were analyzed for VP1 region homology and phylogenetic tree analysis with other isolates at home and abroad. One strain of poliovirus (P1 / 11186), one strain of CV-A4 and one strain of CV-A8, one strain of CV-A2O10 strain CV-B1 / 5 strain CV-B3 and one strain of Echovirusus E (Echovirusus E) type 12, were isolated from the samples of the patients, indicating that there may be an epidemic of enterovirus in the area, and one strain of CV-A4 strain and one strain of CV-A8 strain were isolated from the specimen of the case.
