【摘要】: 目的:探討Neurexin和Neuroligin基因在人類及鼠類動物腸神經系統(tǒng)(ENS)是否有表達,以及對ENS突觸結構與功能的作用,探查其在先天性巨結腸各段的表達特征及意義。 方法:首先制備小鼠、大鼠、豚鼠、正常人體腸管肌間神經叢鋪片,通過雙重免疫熒光細胞染色,證明Neurexin、Neuroligin基因是否在腸神經系統(tǒng)表達,再采用免疫組織化學技術對20例先天性巨結腸狹窄段,移行段和擴張段肌間神經叢鋪片標本染色,判斷Neurexin、Neuroligin基因的表達情況。結果:(1)Neurexin、Neuroligin蛋白在小鼠、大鼠、豚鼠、人體的腸肌間神經叢與神經細胞標記Hu在同一細胞表達;而Neurexin蛋白與突觸前標記物synaptophysin, Neuroligin蛋白與突觸后標記物PSD95也在同一神經組織出現(xiàn)。(2) Neurexin、Neuroligin蛋白在先天性巨結腸擴張段表達最多,移行段明顯減少(P0.05),病變段無表達;同一組織Neurexin和Neuroligin的表達無明顯差異(P>0.05)。 結論:Neurexin、Neuroligin基因在鼠齒類動物及人體腸神經系統(tǒng)都有表達,不僅在胞體,而且在軸突、樹突都有表達,Neurexin在突觸前表達,Neuroligin在突觸后表達。它們對腸神經系統(tǒng)突觸的結構功能起重要作用,與一些胃、腸道動力性疾病有密切關系,是導致先天性巨結腸蠕動功能喪失的重要原因。
[Abstract]:Aim: to investigate the expression of Neurexin and Neuroligin genes in the intestinal nervous system of human and rodent, and to explore the expression characteristics and significance of ENS synaptic structure and function in Hirschsprung's disease (Hirschsprung's disease). Methods: first of all, we prepared mice, rats, guinea pigs and normal human intestinal intermuscular plexus. The expression of Neurexin Neuroligin gene in the enteric nervous system was proved by double immunofluorescence staining. Immunohistochemical staining was used to detect the expression of Neurexin Neuroligin gene in 20 specimens of myenteric plexus of Hirschsprung's disease. Results the expression of Neurexin Neuroligin protein in mouse, rat, guinea pig, human intestinal myenteric plexus and nerve cell labeled Hu was found in the same cell. Neurexin protein and presynaptic marker synaptophysin, Neuroligin protein and postsynaptic marker PSD95 were also found in the same nerve tissue.) Neurexin Neuroligin protein was most expressed in the dilated segment of Hirschsprung's disease. There was no significant difference between the expression of Neurexin and Neuroligin in the same tissue (P > 0.05). Conclusion the expression of Neurexin Neuroligin gene is found in both mouse dentin and human intestinal nervous system. It is expressed not only in the cell body but also in the axons and dendrites. Neurexin is expressed in presynaptic cells and neuroligin is expressed in postsynaptic system. They play an important role in the synaptic structure and function of the intestinal nervous system, and are closely related to some gastric and intestinal dynamic diseases, which are the important reasons for the loss of peristaltic function in Hirschsprung's disease.
