【摘要】： 目的:探討體外條件下不同濃度骨形態(tài)發(fā)生蛋白-2(BMP-2)對脂肪干細(xì)胞誘導(dǎo)分化為成骨細(xì)胞的影響。 方法:本試驗取3周齡雄性SD大鼠,頸部脫臼法處死,無菌條件下取出腹股溝脂肪墊。膠原酶-1消化分離,接種于含有胎牛血清的DMEM培養(yǎng)液中培養(yǎng)。在倒置顯微鏡觀察細(xì)胞形態(tài)及生長情況。待細(xì)胞擴增并生長穩(wěn)定后,分別加8組不同BMP-2濃度(根據(jù)每組BMP-2濃度不同,分為無BMP-2組、100ng/ml BMP-2組、200ng/mlBMP-2組、400ng/ml BMP-2組、800ng/ml BMP-2組、1000ng/mlBMP-2組、1200ng/ml BMP-2組、1600ng/ml BMP-2組)的誘導(dǎo)培養(yǎng)液進(jìn)行成骨細(xì)胞誘導(dǎo)。用堿性磷酸酶、Ⅰ型膠原免疫組織化學(xué)染色及礦化結(jié)節(jié)染色鑒定成骨細(xì)胞分化能力。采用MTT比色法進(jìn)行增殖分化活性比較。 結(jié)果:堿性磷酸酶、Ⅰ型膠原免疫組織化學(xué)染色及礦化結(jié)節(jié)染色均為陽性。與對照組OD均值相比:200ng/ml BMP-2組、400ng/mlBMP-2組、800ng/ml BMP-2組、1200ng/ml BMP-2組、1600ng/mlBMP-2組有顯著性差異(P＜0.05);100ng/ml BMP-2組、1000ng/mlBMP-2組無顯著性差異(P＞0.05)。各組OD值均數(shù)隨BMP-2濃度的增加先逐漸增大(0 ng/ml～400 ng/ml),達(dá)峰值后,又逐漸減少(800 ng/ml～1600 ng/ml)。 結(jié)論:大鼠脂肪組織中分離培養(yǎng)出的脂肪干細(xì)胞(ADSCs)在體外具有向成骨細(xì)胞(OB)分化的潛能。當(dāng)BMP-2濃度在400～800ng/ml時,能促進(jìn)脂肪干細(xì)胞的增殖和分化。
[Abstract]:Aim: to investigate the effects of different concentrations of bone morphogenetic protein-2 (BMP-2) on differentiation of adipose stem cells into osteoblasts in vitro.Methods: male Sprague-Dawley rats aged 3 weeks were killed by cervical dislocations and inguinal fat pads were removed under aseptic condition.Collagenase-1 was digested and isolated and cultured in DMEM medium containing fetal bovine serum.Cell morphology and growth were observed under inverted microscope.The differentiation ability of osteoblasts was evaluated by alkaline phosphatase, type I collagen immunohistochemical staining and mineralized nodule staining.The activity of proliferation and differentiation was compared by MTT colorimetry.Results: alkaline phosphatase, type I collagen immunohistochemical staining and mineralized nodule staining were all positive.With the increase of BMP-2 concentration, the mean OD value of each group increased at first to 0 ng/ml~400 / ml ~ (-1), and then to the peak value, and then decreased to 800 ng/ml~1600 / ml ~ (-1) 路ml ~ (-1) 路L ~ (-1) 路L ~ (-1).Conclusion: adipose stem cells isolated from adipose tissue of rats have the potential to differentiate into osteoblasts in vitro.When the concentration of BMP-2 is 400~800ng/ml, it can promote the proliferation and differentiation of adipose stem cells.
