【摘要】：背景:脂肪干細胞是由中胚層發(fā)育而來的多能干細胞,在特殊的生長因子和環(huán)境等誘導培養(yǎng)條件下,可以向不同的譜系分化。目的:詳細闡述脂肪干細胞誘導分化的條件及鑒定方法。方法:應用計算機檢索萬方數(shù)據(jù)庫及PubM ed數(shù)據(jù)庫2005至2014年10年間的文獻,中文檢索詞為"脂肪干細胞,誘導,分化";英文檢索詞為"adipose derived stem cells,differentiation"。依據(jù)納入排除標準選擇37篇文獻進行歸納總結。結果與結論:脂肪干細胞在抗壞血酸、胰島素、地塞米松、轉化生長因子β作用下可向軟骨細胞分化;成脂誘導液的配方包括3-異丁基-1-甲基黃嘌呤(IBMX)、胰島素、地塞米松、吲哚美辛;成骨分化常用的誘導劑包含地塞米松或維生素D3、抗壞血酸,β-甘油磷酸鈉;堿性成纖維細胞生長因子、表皮生長因子及維生素B27可聯(lián)合應用誘導脂肪干細胞成神經(jīng)分化;向心肌細胞分化普遍應用的誘導因子是5-氮雜胞苷;血管內(nèi)皮生長因子和堿性成纖維細胞生長因子共同作用可以誘導脂肪干細胞向血管內(nèi)皮細胞分化。隨著分子生物學和細胞生物學的迅速發(fā)展,脂肪干細胞的分化研究也會更加深入,在目前對脂肪干細胞誘導分化現(xiàn)象觀察的基礎上,應加強對其內(nèi)在的分子機制及調(diào)控脂肪干細胞可塑性的基因和蛋白的研究。
[Abstract]:Background: adipose stem cells are pluripotent stem cells derived from mesoderm. They are cultured under special conditions such as growth factors and environment. We can differentiate into different lineages. Objective: to elaborate the conditions and identification methods of adipose stem cell induced differentiation. Methods: to search Wanfang database and PubM by computer. Ed database literature for the 10-year period from 2005 to 2014. The key words in Chinese are "adipose stem cells, induction, differentiation"; The key word is "adipose derived stem cells". Differentiation ". Selected 37 articles according to exclusion criteria. Results and conclusions: adipose stem cells in ascorbic acid, insulin, dexamethasone. TGF- 尾 could differentiate into chondrocytes. The formulations of lipopolymers include 3- isobutyl-1-methylxanthine (IBM XN), insulin, dexamethasone, and indomethacin; Common inducers for osteogenic differentiation include dexamethasone or vitamin D _ 3, ascorbic acid, and sodium 尾 -glycerophosphate; Basic fibroblast growth factor, epidermal growth factor and vitamin B27 can be combined to induce neural differentiation of adipose stem cells. 5-azacytidine is the commonly used inducer of cardiomyocyte differentiation. The combination of vascular endothelial growth factor and basic fibroblast growth factor can induce adipose stem cells to differentiate into vascular endothelial cells, with the rapid development of molecular biology and cell biology. The differentiation of adipose stem cells will also be more in-depth, on the basis of the current observation of adipose stem cell induced differentiation. The study of its intrinsic molecular mechanism and genes and proteins regulating the plasticity of adipose stem cells should be strengthened.
