【摘要】: 目的探討新生乳鼠缺血缺氧腦癱模型的制作及評價方法。方法選用出生7天的健康Wistar幼鼠,結(jié)扎右側(cè)頸總動脈結(jié)合8%氧氣、體積分數(shù)92%氮氣環(huán)境缺氧的方法制作幼鼠腦癱模型,不同時間點對實驗動物進行平衡木試驗、足印分析、運動誘發(fā)電位及病理學檢測。結(jié)果(1)足印重復(fù)間距試驗結(jié)果顯示HIE組術(shù)后3周、7周時實驗鼠左側(cè)前后肢足印重復(fù)性差。表現(xiàn)為間距較大,且間距不穩(wěn)定,明顯大于空白對照組(P<0.05);(2)術(shù)后3周、7周平衡木試驗結(jié)果顯示潛伏期及后足滑脫次數(shù)上HIE組明顯多于對照組(P<0.05);(3)術(shù)后3周、7周時HIE組實驗鼠后肢股四頭肌復(fù)合動作電位(CMAP)與對照組相比引出波幅明顯較低(P<0.05);(4)術(shù)后3周,大體觀察右側(cè)腦組織體積變小,萎縮。HE染色光鏡觀察損傷側(cè)腦室周圍細胞腫脹,細胞數(shù)量較對側(cè)少;損傷側(cè)大腦皮層神經(jīng)細胞明顯減少,神經(jīng)細胞變性壞死,膠質(zhì)細胞反應(yīng)性增生。結(jié)論應(yīng)用一側(cè)頸總動脈結(jié)合缺氧的方法制作幼鼠腦癱模型穩(wěn)定、可靠,具有進一步的推廣意義,并可用于腦癱的相關(guān)實驗研究。神經(jīng)行為學及神經(jīng)電生理在腦癱動物模型的評價方面有很大應(yīng)用空間。
[Abstract]:Objective to explore the method of making and evaluating the model of ischemic hypoxic cerebral palsy in neonatal rats. Methods 7 days old healthy Wistar rats, right carotid artery ligation produced cerebral palsy model combined with 8% oxygen, 92% volume fraction of nitrogen oxygen, at different time points of balance beam test, detection of experimental animal footprints analysis, motor evoked potentials and pathology. Results (1) the results of repeat interval test showed that the left hind limbs of the HIE group had poor reproducibility at the left and the left hind limbs at 3 and 7 weeks after operation. For larger spacing and the spacing is not stable, significantly greater than the control group (P < 0.05); (2) after 3 weeks, 7 weeks, balance beam test results show that the latency and the number of foot slippage in HIE group was higher than that of control group (P < 0.05); (3) 3 weeks after surgery, 7 week HIE group experimental rat femoral head four muscle compound action potential (CMAP) compared with the control group leads to amplitude was significantly lower (P < 0.05); (4) 3 weeks after operation, gross observation of the right side of the brain tissue volume, atrophy. HE staining and light microscopy showed that the cells in the injured side ventricles were swollen, the number of cells was less than that on the contralateral side, the neurons in the injured side were significantly reduced, the degeneration and necrosis of neurons, and the reactive proliferation of glial cells. Conclusion it is stable and reliable to use unilateral common carotid artery combined with anoxia to make cerebral palsy model in young rats, which has further popularization significance and can be used for experimental study of cerebral palsy. Neurobehavioral and neuroelectrophysiology are very useful in the evaluation of cerebral palsy animal models.
