Dysbindin在人和實驗動物顳葉癲癇灶中的表達及意義

目的:Dysbindin蛋白在中樞神經(jīng)系統(tǒng)的多個部位廣泛表達,它在細胞內(nèi)囊泡轉運、突觸囊泡轉運和神經(jīng)遞質(zhì)釋放等方面發(fā)揮重要的作用。在細胞水平,Dysbindin被認為調(diào)節(jié)突觸前的谷氨酸能傳遞。大腦中興奮性的和抑制性的神經(jīng)遞質(zhì)之間出現(xiàn)平衡失調(diào)密切關系到癲癇的發(fā)生。而谷氨酸是大腦中含量最為豐富的一種興奮性神經(jīng)遞質(zhì),并且在癲癇活動的啟動和擴散中發(fā)揮作用。本課題旨在研究Dysbindin在顳葉癲癇患者和氯化鋰-匹羅卡品癲癇大鼠模型腦組織中的表達變化,探討Dysbindin與癲癇發(fā)生的關系。方法:在前期從耐藥性顳葉癲癇患者中收集的腦組織庫中隨機選取25例患者的顳葉皮質(zhì)和10例對照顳葉組織,同時用氯化鋰-匹羅卡品誘導建立顳葉癲癇模型,收集大鼠海馬組織和鄰近皮質(zhì)。運用免疫熒光和western blot的方法檢測Dysbindin表達的部位及變化規(guī)律。結果:在人顳葉組織中,Dysbindin在神經(jīng)元胞漿中表達,其在顳葉癲癇患者中的表達水平較對照組顯著增高。同樣,Dysbindin在大鼠的實驗組和對照組的海馬和鄰近皮質(zhì)的神經(jīng)元中表達。大鼠腦組織的western blotting結果顯示:Dysbindi...
