Notch信號(hào)通路在大鼠骨髓間充質(zhì)干細(xì)胞移植后促進(jìn)腦缺血區(qū)血管新生過(guò)程中的作用

【摘要】:目的探討Notch1信號(hào)通路在骨髓間充質(zhì)干細(xì)胞(MSCs)移植后促進(jìn)腦缺血區(qū)血管新生過(guò)程中的作用。方法分離、培養(yǎng)SD大鼠MSCs,制作大鼠大腦中動(dòng)脈栓塞(MCAO)模型,將試驗(yàn)分為正常組、腦缺血后MSCs移植組、腦缺血后MSCs移植+rh NF-κB組、腦缺血對(duì)照組。腦組織冰凍切片行Ⅷ因子免疫組織熒光染色檢測(cè)各試驗(yàn)組MSCs移植1、4、7、14 d后缺血皮質(zhì)區(qū)微血管數(shù)量。Western印跡檢測(cè)各試驗(yàn)組MSCs移植1、4、7、14 d后缺血皮質(zhì)區(qū)血管內(nèi)皮生長(zhǎng)因子(VEGF)165、Notch1、Hes1蛋白表達(dá)。結(jié)果缺血對(duì)照組、MSCs移植組和MSCs移植+rh NF-κB組在各時(shí)間點(diǎn)微血管數(shù)量及VEGF165、Notch1、Hes1蛋白表達(dá)均顯著高于正常組,從第14天起開始下降,3組的微血管數(shù)量及VEGF165、Notch1、Hes1蛋白表達(dá)依次上升,差異顯著(P0.01)。結(jié)論 MSCs移植可促進(jìn)缺血區(qū)新生血管的形成,并可通過(guò)激活Notch信號(hào)通路促進(jìn)缺血皮質(zhì)區(qū)VEGF165的表達(dá),從而促進(jìn)缺血區(qū)的血管新生。
