酒精作用下小鼠海馬區(qū)神經(jīng)元放電規(guī)律研究

【摘要】:慢性和急性飲酒均可引起腦損傷,目前對急性飲酒導(dǎo)致的大腦記憶的損傷研究多采用生物化學(xué)及行為學(xué)方法。本文利用微電極陣列系統(tǒng)記錄活體小鼠海馬區(qū)神經(jīng)元放電電位,分析并獲得腦神經(jīng)元放電規(guī)律,研究急性飲酒對小鼠記憶的影響。進(jìn)一步通過激光散斑襯比成像實(shí)驗(yàn)和小鼠行為學(xué)T型迷宮實(shí)驗(yàn),驗(yàn)證了神經(jīng)元放電實(shí)驗(yàn)結(jié)果的正確性。論文主要研究內(nèi)容包括:1、闡述了大腦神經(jīng)元電信號(hào)與腦記憶生理特征的內(nèi)在聯(lián)系,以及神經(jīng)元電生理技術(shù)在探討急性飲酒對小鼠記憶影響方面的研究進(jìn)展;2、編寫了基于Matlab的信號(hào)預(yù)處理程序,實(shí)現(xiàn)了小鼠大腦海馬CA1區(qū)的神經(jīng)元放電信號(hào)的噪聲濾除、鋒電位特征提取與聚類;3、設(shè)計(jì)并完成了急性飲酒(重度、輕度)下小鼠的電生理實(shí)驗(yàn),采集并分析了小鼠大腦CA1區(qū)鋒電位的變化規(guī)律;另外,采用激光散斑襯比成像技術(shù)測量了腦局部血、利用T型迷宮研究了急性飲酒小鼠的方向選擇正確率,驗(yàn)證了電生理實(shí)驗(yàn)結(jié)果;4、設(shè)計(jì)實(shí)驗(yàn)進(jìn)一步研究了常用解酒藥物葛根素對急性飲酒小鼠神經(jīng)元放電規(guī)律的影響,研究了葛根素對大腦記憶的保護(hù)作用。論文采用多通道微電極陣列系統(tǒng)采集了急性飲酒小鼠大腦CA1區(qū)神經(jīng)元鋒電位,結(jié)果表明急性重度飲酒對小鼠海馬區(qū)神經(jīng)元放電具有抑制作用,急性輕度飲酒對海馬區(qū)神經(jīng)元放電具有促進(jìn)作用。同時(shí),葛根素對急性飲酒引起的神經(jīng)元放電變化具有拮抗作用,證明了葛根素對飲酒機(jī)體大腦的保護(hù)作用。腦局部血流實(shí)驗(yàn)和行為學(xué)實(shí)驗(yàn)充分驗(yàn)證了電生理實(shí)驗(yàn)結(jié)果的正確性。論文的研究結(jié)論在探究急性飲酒對小鼠記憶的影響及葛根素對急性飲酒造成的大腦損傷的保護(hù)作用中具有重要的理論意義和臨床應(yīng)用價(jià)值。
[Abstract]:Both chronic and acute drinking can cause brain damage. At present, biochemical and behavioral methods are used to study the brain memory impairment caused by acute drinking. In this paper, the discharges of neurons in the hippocampal area of living mice were recorded by microelectrode array system, and the discharge patterns of brain neurons were analyzed and obtained, and the effects of acute drinking on the memory of mice were studied. The experimental results of firing of neurons were verified by laser speckle contrast imaging and behavioral T-maze test in mice. The main contents of this paper include: 1, the intrinsic relationship between the electrical signals of brain neurons and the physiological characteristics of brain memory. The research progress of neuron electrophysiology in the study of the effect of acute drinking on the memory of mice was also discussed. A signal preprocessing program based on Matlab was written to remove the noise of the neuron discharge signal in the CA1 region of the hippocampus of the mouse brain. The electrophysiological experiments of mice under acute drinking (severe and mild) were designed and completed. The changes of spike potentials in CA1 region of mice were collected and analyzed. The regional blood of brain was measured by laser speckle contrast imaging, and the correct rate of direction selection in acute drinking mice was studied by using T-maze. The experiment was designed to study the effect of puerarin, a common antialcoholic drug, on the firing of neurons in acute drinking mice and the protective effect of puerarin on brain memory. Multichannel microelectrode array system was used to collect neuronal spike potentials in CA1 area of acute drinking mice. The results showed that acute severe drinking could inhibit neuronal discharges in hippocampus of mice. Acute mild alcohol consumption can promote neuronal discharges in the hippocampal area. At the same time, puerarin has antagonistic effect on the changes of neuronal discharges induced by acute drinking, which proves the protective effect of puerarin on the brain of drinking body. The experimental results of local cerebral blood flow and behavior fully verify the correctness of the results of electrophysiological experiments. The conclusion of this paper has important theoretical significance and clinical application value in exploring the effect of acute drinking on the memory of mice and the protective effect of puerarin on brain injury caused by acute drinking.
