【摘要】：目的觀察綠茶提取物EGCG對實驗性自身免疫性腦脊髓炎(EAE)小鼠的治療作用及相關機制。方法取C57BL/6小鼠,隨機分為對照組、EAE模型組和EGCG治療組,對各組小鼠進行神經(jīng)功能評分,并對中樞神經(jīng)系統(tǒng)炎性浸潤情況進行觀察;流式細胞技術檢測Th1、Th17細胞在中樞神經(jīng)系統(tǒng)中頻率變化;免疫組化觀察25 d腦組織IFN-γ、IL-17和TGF-β的表達情況;ELISA檢測血清中IFN-γ、、IL-17、TGF-β、IL-10、IL-4的質量濃度。結果 EGCG治療明顯緩解EAE小鼠疾病癥狀和中樞神經(jīng)系統(tǒng)炎癥,抑制Th1與Th17細胞向中樞神經(jīng)系統(tǒng)浸潤。與EAE模型組相比,EGCG治療組腦IFN-γ及IL-17表達上調,而TGF-β表達下調;外周血中可溶性上述細胞因子表達情況與腦一致。結論 EGCG可能通過影響中樞神經(jīng)系統(tǒng)及外周血中IFN-γ、IL-17、TGF-β的表達,調控T細胞亞群的分化對EAE發(fā)揮保護作用。
[Abstract]:Objective to observe the therapeutic effect of green tea extract (EGCG) on experimental autoimmune encephalomyelitis (EAE) mice and its related mechanism. Methods C57BL/6 mice were randomly divided into two groups: the control group and the EGCG group. The neurological function of each group was evaluated and the inflammatory infiltration of the central nervous system was observed. The expression of IFN- 緯 IL-17 and TGF- 尾 in human brain tissue was detected by flow cytometry and the mass concentration of IL-10 IL-4 in serum was detected by Elisa. Results EGCG significantly alleviated the disease symptoms and central nervous system inflammation in EAE mice and inhibited the infiltration of Th1 and Th17 cells into the central nervous system. Compared with the EAE model group, the expression of IFN- 緯 and IL-17 was up-regulated, but the expression of TGF- 尾 was down-regulated, and the expression of soluble cytokines in peripheral blood was consistent with that in the brain. Conclusion EGCG may play a protective role on EAE by affecting the expression of IFN- 緯 IL-17 TGF- 尾 in the central nervous system and peripheral blood and regulating the differentiation of T cell subsets.
