【摘要】：目的研究鹽酸法舒地爾對蛛網(wǎng)膜下腔出血(SAH)大鼠學習記憶及海馬CA1區(qū)自噬的影響。方法雄性Sprague-Dawley大鼠54只,隨機分為假手術組、SAH組、藥物組,各18只。頸內(nèi)動脈刺破法造模,藥物組造模成功后予鹽酸法舒地爾10 mg/kg腹腔注射,每天1次,假手術組和SAH組腹腔注射等量生理鹽水,干預后6 h、24 h、72 h予穿梭箱試驗;取海馬CA1區(qū)組織HE染色觀察細胞形態(tài),免疫組織化學染色觀察Beclin-1和微管相關蛋白l輕鏈3-Ⅱ(LC3-Ⅱ)的表達。結果與假手術組相比,SAH組大鼠各時間點穿梭箱試驗逃避反應次數(shù)減少(P0.05),逃避反應時間延長(P0.05);海馬CA1區(qū)神經(jīng)細胞數(shù)目減少(P0.05),Beclin-1、LC3-Ⅱ表達增高(P0.05)。與SAH組相比,藥物組各時間點逃避反應次數(shù)增多(P0.05),逃避反應時間減少(P0.05);海馬CA1區(qū)神經(jīng)細胞增多(P0.05),Beclin-1、LC3-Ⅱ表達進一步增多(P0.05)。結論鹽酸法舒地爾可改善SAH大鼠學習記憶功能,減少神經(jīng)細胞損傷,增強海馬CA1區(qū)自噬。
[Abstract]:Objective to study the effects of fasudil hydrochloride on learning and memory and hippocampal CA1 autophagy in rats with subarachnoid hemorrhage (SAH). Methods 54 male Sprague-Dawley rats were randomly divided into sham-operated group (n = 18) and drug group (n = 18). The internal carotid artery puncture method was used to make the model, and the drug group was injected intraperitoneally with fasudil hydrochloride for 10 mg/kg once a day. The sham operation group and SAH group were injected with the same amount of normal saline intraperitoneally.
