【摘要】：目的檢測(cè)GBM患者和健康人外周血中miRNA的異常表達(dá)。方法利用基因芯片分析GBM患者和正常對(duì)照者血清中miRNA的表達(dá)量,然后進(jìn)一步進(jìn)行靶基因的生物信息學(xué)分析。結(jié)果miRNA芯片表明在血清中15例GBM組和15例正常對(duì)照組的外周血miRNA表達(dá)有明顯差異。752種miRNA中,GBM組有95種miRNA表達(dá)上調(diào),22種表達(dá)下調(diào)。(倍數(shù)≥2.0,P0.01)。通過(guò)深入分析,我們發(fā)現(xiàn)GBM患者miR-576-5p、miR-340和miR-626過(guò)表達(dá),但miR-320、let-7g-5p、miR-7-5p低表達(dá)。進(jìn)一步相關(guān)生物信息學(xué)分析,我們發(fā)現(xiàn),他們?cè)谀X膠質(zhì)瘤信號(hào)通路的調(diào)控中發(fā)揮重要的作用。結(jié)論與正常人比較,以上6種miRNA在GBM患者外周血中有顯著差異。外周血中的miRNA表達(dá)譜中一些特異性表達(dá)的miRNA可能作為高特異性和靈敏度診斷膠質(zhì)瘤的新靶向標(biāo)志物。
[Abstract]:Objective to detect the abnormal expression of miRNA in peripheral blood of patients with GBM and healthy people. Methods the expression of miRNA in serum of GBM patients and normal controls was analyzed by gene microarray, and then the bioinformatics analysis of target genes was carried out. Results miRNA microarray showed that there were significant differences in the expression of miRNA in peripheral blood between 15 cases of GBM group and 15 cases of normal control group.
