【摘要】：目的研究負(fù)載癌-睪丸抗原(cancer-testis antigen,CTA)OY-TES-1的樹突狀細(xì)胞(dendritic cell,DC)體外抗膠質(zhì)瘤的細(xì)胞免疫效應(yīng)。方法細(xì)胞因子聯(lián)合誘導(dǎo)HLA-A2+健康人外周血單核細(xì)胞為DC,采用OY-TES-1重組蛋白致敏DC,流式細(xì)胞術(shù)檢測成熟DC表型分子;進(jìn)而使DC誘導(dǎo)同體T淋巴細(xì)胞產(chǎn)生細(xì)胞毒性T淋巴細(xì)胞(cytotoxic T lymphocyte,CTL),CCK8檢測T細(xì)胞增殖情況,ELISPOT檢測IFN-γ含量,乳酸脫氫酶釋放法檢測CTL對膠質(zhì)瘤細(xì)胞U251和A172的殺傷作用。結(jié)果負(fù)載OY-TES-1的DC低表達(dá)CD14,而高表達(dá)CD1a、CD83和CD80表型分子,能明顯促進(jìn)CD8+T細(xì)胞的增殖及其IFN-γ的分泌(P0.05);而OY-TES-1特異性CTL在效靶比為50∶1時(shí)對HLA-A2+和OY-TES-1+U251、HLA-A2-和OY-TES-1+A172均有明顯殺傷作用(均P0.05),但其對U251的殺傷作用更強(qiáng)。結(jié)論體外用OY-TES-1重組蛋白致敏的DC可誘導(dǎo)特異性抗膠質(zhì)瘤免疫應(yīng)答,提示OY-TES-1可能成為膠質(zhì)瘤免疫治療的靶點(diǎn)。
[Abstract]:Objective to study the effect of dendritic cells loaded with cancer-testis antigen1 (CTAOY-TES-1) on glioma in vitro. Methods HLA-A2 monocytes were induced by cytokines and OY-TES-1 recombinant protein was used to sensitize DC. Flow cytometry was used to detect the phenotypic molecules of mature DC. The cytotoxic T lymphocytes were induced by DC to produce cytotoxic T lymphocytes. The proliferation of T cells was detected by CCK8 and IFN- 緯 by Elispot. The cytotoxicity of CTL on U251 and A172 glioma cells was detected by lactate dehydrogenase release assay. Results the expression of CD14 was low in DC loaded with OY-TES-1, while the phenotypic molecules of CD1a, CD83 and CD80 were highly expressed in DC. It could obviously promote the proliferation of CD8 T cells and the secretion of IFN- 緯, while OY-TES-1 specific CTL could kill both HLA-A2 and OY-TES-1 U251 HLA-A2- and OY-TES-1 A172 at 50:1. Conclusion DC sensitized with OY-TES-1 recombinant protein in vitro can induce specific anti-glioma immune response, suggesting that OY-TES-1 may be the target of glioma immunotherapy.
