失肌皮神經(jīng)與屈肘重建后供受體肌肌肉型AChR亞型表達(dá)變化

【摘要】:目的:探索失肌皮神經(jīng)損傷和屈肘重建術(shù)后,肱二頭。ㄊ荏w。┖托卮蠹⌒辈浚üw。﹥(nèi)肌肉型AChR亞型(ε-AChR和γ-AChR)的表達(dá)變化規(guī)律,為防治供受體肌的萎縮提供科學(xué)指導(dǎo)。 方法:1.實(shí)驗(yàn)動(dòng)物及分組:42只成年SD大鼠,體重200±50g,不拘雌雄,分A、B、C、D4組,,其中B、C、D組又分別分為1、2組;它們分別是A組(正常組),B1、B2組(肌皮神經(jīng)切斷2、4w組),C1、C2組(假移植術(shù)2、4w組),D1、D2組(重建屈肘功能術(shù)2、4w組),每組6只。2.大體解剖。3.建立失肌皮神經(jīng)損傷后胸大肌斜部移位重建屈肘功能動(dòng)物模型。4.肌形態(tài)學(xué)觀察和稱量肌濕重。5. RT-PCR檢測(cè)ε-AChR和γ-AChR mRNA表達(dá)。6. Western blot檢測(cè)ε-AChR和γ-AChR蛋白表達(dá)。 結(jié)果:1.大體觀察:胸大肌斜部移位縫合至肱二頭肌腱止端時(shí),不會(huì)傷及血管神經(jīng),能滿足無張力縫合。失肌皮神經(jīng)后肱二頭肌進(jìn)行性萎縮、色澤變淡、覓食受限;重建屈肘術(shù)后,上述變化逐漸改善,胸大肌斜部手術(shù)前后變化不大。2.肌濕重:肱二頭肌A組濕重為0.38±0.01g,肌皮神經(jīng)切斷后,B組和C組與A組比較,進(jìn)行性減輕;屈肘重建后,D組與A組比較,逐漸回升,p<0.05。胸大肌斜部A組肌重0.37±0.01g,屈肘重建后,D1組與A組比較,下降2.70%,p<0.05;D2組已回升,與A組比較,p>0.05。3. mRNA表達(dá)水平:肱二頭肌和胸大肌斜部A組內(nèi)ε-AChR表達(dá)水平分別為0.66±0.03和0.82±0.02,γ-AChR無表達(dá)。肱二頭肌內(nèi)ε-AChR的表達(dá),B、C組與A組比較,逐漸下降,γ-AChR重新表達(dá)并逐漸上調(diào);重建屈肘術(shù)后,D組與B2組比較,ε-AChR表達(dá)明顯上調(diào),γ-AChR下調(diào),C組與B2組比較,結(jié)果相反,p<0.05。胸大肌斜部?jī)?nèi)ε-AChR表達(dá),D1和D2組與A組比較,分別下降了14.63%、1.22%,D1組與A組比較,p<0.05,但D2組與A組比較,p>0.05;γ-AChR在D1組重新表達(dá)為0.18±0.01,D2組幾乎消退,僅為0.01±0.01,D2組和D1組比較,下調(diào)明顯,p<0.05。4.蛋白表達(dá):各組各時(shí)間點(diǎn)蛋白表達(dá)變化規(guī)律與mRNA表達(dá)相同,除A組外,各組各時(shí)間點(diǎn)的蛋白表達(dá)變化幅度均低于mRNA的表達(dá)。 結(jié)論:1. SD大鼠胸大肌斜部作為供體肌,建立屈肘重建術(shù)動(dòng)物模型可行,能改善肱二頭。ㄊ荏w。┪s;2.失肌皮神經(jīng)后,肱二頭肌內(nèi)ε-AChR mRNA和蛋白表達(dá)逐漸下調(diào),而γ-AChR重新表達(dá)并逐漸上調(diào);屈肘重建術(shù)后,受體肌內(nèi)這兩個(gè)受體亞型的表達(dá)變化相反;3.該屈肘重建術(shù),供體肌內(nèi)γ和ε-AChR僅一過性異常表達(dá)。
[Abstract]:Objective: to investigate the expression of AChR subtypes (蔚 -AChR and 緯 -AChR) in the biceps brachii (recipient muscle) and the oblique pectoralis major (donor muscle) after denervated cutaneous nerve injury and elbow flexion reconstruction, so as to provide scientific guidance for the prevention and treatment of donor muscle atrophy. Methods: 1. The experimental animals and 42 adult SD rats were divided into two groups. The weight was 200 鹵50 g. The rats were divided into two groups. They were group A (normal group), group B _ (1) B _ (B _ (2)) (group C _ (1) C _ (1) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _ (2) C _. Observation of muscle morphology and weighing of wet muscle weight. RT-PCR was used to detect the expression of 蔚 -AChR and 緯 -AChR mRNA. 6. Western blot was used to detect the expression of 蔚 -AChR and 緯 -AChR protein. Results 1. General observation: when the oblique part of pectoralis major muscle was sutured to the end of biceps brachii tendon, it would not injure the blood vessel and nerve, and could satisfy the tension-free suture. The biceps biceps after the loss of musculocutaneous nerve gradually atrophied, the color became light, and the foraging was limited. After elbow flexion reconstruction, the above changes were gradually improved. The changes of muscle wet weight were not significant before and after operation. The wet weight of biceps brachii muscle group A was 0.38 鹵0.01g, and that of group B and C decreased progressively compared with group A after transection of musculocutaneous nerve. The muscle weight of group A was 0.37 鹵0.01 g in oblique part of pectoralis major, and decreased 2.70% (P < 0.05) in group D after elbow flexion reconstruction compared with group A. Compared with group A, the expression level of 蔚 -AChR in biceps brachii muscle and oblique part of pectoralis major muscle was 0.66 鹵0.03 and 0.82 鹵0.02respectively, but the expression of 蔚 -AChR in biceps brachii muscle was not expressed. The expression of 蔚 -AChR in biceps brachii muscle was decreased gradually compared with group A, but the expression of 緯 -AChR was gradually up-regulated. Compared with group B2, the expression of 蔚 -AChR in group D was significantly up-regulated, and the expression of 蔚 -AChR in group D was significantly higher than that in group B2 (P < 0.05). The expression of 蔚 -AChR in the oblique part of pectoralis major was significantly higher in group D than that in group B 2, and the expression of 蔚 -AChR in group D was significantly higher than that in group A (P < 0.05). The expression of 緯 -AChR in D1 group was almost disappeared to 0.18 鹵0.01D 2 group, only 0.01 鹵0.01 渭 g D 2 group, but P > 0.05 in D 2 group compared with A group, and the expression of 緯 -AChR in D 1 group was less than that in A group (P < 0.05), and the expression of 緯 -AChR in D 1 group was almost disappeared, only 0.01 鹵0.01 渭 g D 2 group and D 1 group, P < 0.05, P < 0.05, P < 0.05, respectively. The expression of protein was decreased significantly (P < 0.05.4). The change of protein expression at each time point in each group was the same as that in mRNA, except for group A, the change range of protein expression at each time point in each group was lower than that of mRNA expression. Conclusion 1. The oblique part of the pectoralis major muscle of SD rats is feasible to establish the animal model of elbow flexion reconstruction, which can improve the atrophy of the biceps brachii (receptor muscle). 2. The expression of 蔚 -AChR mRNA and protein in the biceps brachii is gradually down-regulated after the loss of the cutaneous nerve of the biceps brachii. After elbow flexion reconstruction, the expression of these two receptor subtypes was reversed. 3. In this elbow flexion reconstruction, the expression of 緯 and 蔚 -AChR in donor muscle was only transient abnormal.
