扇貝裙邊糖胺聚糖對(duì)六羥基多巴胺誘導(dǎo)SH-SY5Y細(xì)胞損傷的保護(hù)作用研究

【摘要】:目的:研究扇貝裙邊提取物—扇貝糖胺聚糖(SS-GAG)對(duì)六羥基多巴胺(6-OHDA)誘導(dǎo)的多巴胺能神經(jīng)元細(xì)胞損傷的保護(hù)作用,及其抗帕金森病(parkinson's disease PD)的作用機(jī)制。 方法:體外培養(yǎng)人神經(jīng)母瘤細(xì)胞株SH-SY5Y,隨機(jī)分為對(duì)照組、模型組(六羥基多巴胺損傷組)和實(shí)驗(yàn)組(不同濃度的SS-GAG預(yù)處理細(xì)胞后,再加入六羥基多巴胺),采用MTT法測(cè)定細(xì)胞的存活率,Hochest33258熒光染色觀察凋亡細(xì)胞的形態(tài)學(xué)變化,Annexin V-FITC/PI雙染法檢測(cè)細(xì)胞凋亡率,流式細(xì)胞術(shù)檢測(cè)線粒體膜電位的改變。 結(jié)果:六羥基多巴胺損傷細(xì)胞后,細(xì)胞存活率顯著降低;熒光顯微鏡下觀察細(xì)胞出現(xiàn)典型的凋亡形態(tài);細(xì)胞凋亡率顯著升高;線粒體膜電位顯著下降。而用SS-GAG預(yù)處理24小時(shí)后,能顯著提高細(xì)胞存活率;細(xì)胞凋亡程度明顯減弱,凋亡率有明顯下降;升高線粒體膜電位。結(jié)論:SS-GAG能夠減輕六羥基多巴胺對(duì)SH-SY5Y細(xì)胞的損傷程度,且在一定濃度范圍內(nèi)呈劑量依賴效應(yīng)。
[Abstract]:Objective: To study the protective effect of scallop skirt extract scallop glycosaminoglycan (SS-GAG) on six hydroxydopamine (6-OHDA) induced dopaminergic neuron cell injury and its mechanism of anti Parkinson's disease (parkinson's disease PD).
Methods: human neuroblastoma cell line SH-SY5Y cultured in vitro were randomly divided into control group, model group (injury group six hydroxy dopamine) and experimental group (pretreatment with various concentrations of SS-GAG cells after adding six Ohda), the survival rate of cells was determined by MTT assay, morphological changes of apoptosis was observed by Hochest33258 fluorescence Annexin V-FITC/PI staining method to detect cell apoptosis rate, mitochondrial membrane potential detection of flow cytometry.
Results: six hydroxydopamine damage cells, cell survival rate decreased significantly; under fluorescence microscope the cells appeared typical apoptotic morphology; the apoptosis rate was significantly increased; mitochondrial membrane potential decreased significantly. While pretreatment with SS-GAG after 24 hours, can significantly increase the cell survival rate; cell apoptosis significantly reduced, apoptosis rate the mitochondrial membrane potential decreased significantly; increased. Conclusion: SS-GAG can reduce six hydroxydopamine injury on SH-SY5Y cells in a dose-dependent manner, and in a certain concentration range.
