【摘要】：目的：研究人腦視皮質(zhì)中PV、NeuN陽(yáng)性神經(jīng)元隨著衰老而發(fā)生的改變,為研究退行性病變提供形態(tài)學(xué)依據(jù)。 方法：(1)收集22-102歲人腦標(biāo)本17例。取視皮質(zhì),固定,切片,免疫組織化學(xué)方法染色視皮質(zhì)PV、NeuN陽(yáng)性細(xì)胞,光鏡下計(jì)算細(xì)胞密度和光密度值,統(tǒng)計(jì)分析。(2)對(duì)上述17例視皮質(zhì)鄰片進(jìn)行6E10免疫組織化學(xué)染色,并分析6E10與PV, NeuN的相關(guān)性。 結(jié)果：1.視皮質(zhì)內(nèi)PV陽(yáng)性神經(jīng)元主要分布在外顆粒層、外錐體細(xì)胞層和內(nèi)顆粒層。細(xì)胞形態(tài)大小不一,胞體呈卵圓形,梭形或者不規(guī)則形,突起清晰可辨。PV陽(yáng)性細(xì)胞數(shù)目隨衰老而逐漸下降。2.NeuN陽(yáng)性神經(jīng)元分布于視皮質(zhì)的各層,胞核深染,胞漿和樹(shù)突淡染。細(xì)胞隨衰老而出現(xiàn)密度下降、體積變大、空泡化等現(xiàn)象。3.有6E10免疫陽(yáng)性產(chǎn)物的視皮質(zhì),其PV和NeuN免疫陽(yáng)性神經(jīng)元數(shù)量下降明顯。 結(jié)論：人視皮質(zhì)PV和NeuN免疫陽(yáng)性神經(jīng)元數(shù)目隨衰老而下降,有6E10免疫陽(yáng)性斑塊出現(xiàn)的標(biāo)本,其數(shù)目下降更為明顯。
[Abstract]:Aim: to study the changes of PVN Neun positive neurons in the visual cortex of human brain with aging, and to provide morphological evidence for the study of degenerative lesions. Methods 17 human brain specimens aged 22 to 102 years were collected. The visual cortex was fixed, sectioned and stained with immunohistochemical staining for PVN positive cells in the visual cortex. The cell density and optical density were calculated under light microscope. Statistical analysis. (2) 6E10 immunohistochemical staining was performed on the adjacent sections of the visual cortex of 17 cases mentioned above, and 6E10 and PV were analyzed. Correlation of NeuN. Results 1. PV-positive neurons in the visual cortex were mainly distributed in the outer granular layer, the outer pyramidal cell layer and the inner granular layer. The size of the cells varied and the cell bodies were oval, fusiform or irregular. The number of clearly identifiable. PV positive cells decreased with aging. 2. Neun positive neurons were distributed in all layers of the visual cortex and the nucleus was deeply stained. Light staining of cytoplasm and dendrites. The density of cells decreased with aging, the volume became larger, vacuolation and other phenomena. 3. There were 6E10 immunoreactive visual cortex. The number of PV and NeuN immunoreactive neurons decreased significantly. Conclusion: the number of PV and NeuN immunoreactive neurons in human visual cortex decreases with aging, and the number of 6E10 immunoreactive plaques decreases more obviously.
