神經(jīng)酸對(duì)帕金森病模型小鼠運(yùn)動(dòng)障礙的緩解作用及機(jī)制研究

【摘要】:目的:研究神經(jīng)酸對(duì)帕金森病(PD)模型小鼠運(yùn)動(dòng)障礙的緩解作用及機(jī)制。方法:將小鼠隨機(jī)分為空白對(duì)照組(生理鹽水)、模型組(生理鹽水)、多巴絲肼片組(陽性對(duì)照,按左旋多巴計(jì)120 mg/kg)和神經(jīng)酸低、中、高劑量組(20.0、40.0、80.0 mg/kg),每組10只。除空白對(duì)照組外,其余各組小鼠均建立PD模型。成模后,每天ig相應(yīng)藥物1次,連續(xù)14 d。末次給藥后,觀察各組小鼠行為學(xué)變化,采用高效液相色譜法測(cè)定小鼠腦紋狀體內(nèi)多巴胺(DA)及其代謝物二羥苯乙酸(DOPAC)、高香草酸(HVA)的含量。結(jié)果:與空白對(duì)照組比較,模型組小鼠爬桿時(shí)間延長(zhǎng)、滾筒時(shí)間縮短、自發(fā)運(yùn)動(dòng)次數(shù)減少(P0.05),腦紋狀體內(nèi)DA、DOPAC、HVA含量均降低(P0.05)。與模型組比較,多巴絲肼片組和神經(jīng)酸各劑量組小鼠爬桿時(shí)間縮短、滾筒時(shí)間延長(zhǎng)(P0.05),腦紋狀體內(nèi)DA、DOPAC、HVA含量均升高(P0.05);多巴絲肼片組和神經(jīng)酸高劑量組小鼠自發(fā)運(yùn)動(dòng)次數(shù)增加(P0.05)。結(jié)論:神經(jīng)酸可有效改善PD模型小鼠的運(yùn)動(dòng)障礙癥狀,其機(jī)制可能與增加腦紋狀體內(nèi)DA含量有關(guān)。
[Abstract]:Objective: to study the effect and mechanism of nerve acid on motor disorder in PD mice. Methods: the mice were randomly divided into control group (normal saline group) and model group (normal saline group). Dobutylhydrazine group (positive control, 120 mg / kg L-dopa) and low, high dose group (20.0 ~ 40.0 mg 路kg ~ (-1)). PD model was established in all mice except the blank control group. After the model was established, the mice were given the corresponding drugs once a day for 14 days, and the behavioral changes of each group were observed. High performance liquid chromatography (HPLC) was used to determine the contents of dopaminergic acid (DAA) and its metabolites (DOPAC, HVA) in the striatum of mice. Results: compared with the blank control group, the contents of dopamine (DA) and dihydroxyphenylacetate (DOPA) were determined by high performance liquid chromatography. In the model group, the time of climbing rod was prolonged, the time of roller was shortened, the number of spontaneous movement was decreased (P 0.05), and the DAA DOPAC in the striatum was decreased. Compared with the model group, compared with the model group, the mice in the dobutazide group and the nerve acid group had shorter climbing time, longer roller time and DA in the striatum. The content of HVA in DOPAC was increased (P 0.05). The number of spontaneous exercise increased in dobutylhydrazine group and high dose group. Conclusion: neuronal acid can effectively improve the symptoms of motor disorder in PD model mice. The mechanism may be related to the increase of DA content in the striatum.
