1320nmND:YAG激光治療皮膚光老化的動物實驗研究

【摘要】:背景與目的 皮膚光老化主要指人的皮膚受紫外線等外界環(huán)境因素的影響出現(xiàn)衰老的現(xiàn)象,主要表現(xiàn)為皮膚粗糙、皺紋色斑出現(xiàn)等,不僅影響一個人的外表,而且與皮膚癌有正相關性,它的發(fā)病機制目前不完全清楚。皮膚光老化的治療方法較多,YAG激光治療是近幾年發(fā)展起來的除皺緊膚術,具有創(chuàng)傷小,臨床愈合快的特點,在臨床上治療皮膚光老化取得了不錯的療效,但其具體治療機制目前并不清楚。本研究通過大鼠皮膚光老化動物模型,進一步研究皮膚光老化和YAG激光治療的機制,以期對皮膚光老化的臨床治療有所幫助。 方法 選取雄性SD大鼠40只作為研究對象,隨機選取10只作為對照組,其余30只復制皮膚光老化模型,模型復制成功后再隨機分成三組(光老化組、激光治療組、自然恢復組),每只大鼠皮膚處理部位分別做組織病理HE染色,并提取RNA,逆轉錄成cDNA,做SYBR Green Ⅰ real time PCR檢測四組大鼠皮膚中β-actin、Ⅰ型膠原、Ⅲ型膠原、CTGF、TGF-β1的mRNA表達情況。 結果 肉眼觀察:光老化組大鼠背部皮膚增厚,粗糙,出現(xiàn)較深皺紋,經(jīng)YAG激光治療后大鼠皮膚光老化癥狀減輕,而自然恢復組變化不明顯;顯微鏡觀察:光老化組大鼠皮膚HE染色,表皮紊亂,疏松增厚、厚薄不均,真皮膠原纖維斷裂、排列扭曲紊亂,有炎性細胞浸潤,YAG激光治療后,癥狀明顯改善;與對照組相比,光老化組Ⅰ型膠原、Ⅲ型膠原、CTGF、TGF-β1mRNA表達明顯降低,YAG激光治療后,這些基因mRNA表達明顯升高,激光治療組與自然恢復組相比,除CTGF外,Ⅰ型膠原、Ⅲ型膠原、TGF-p的mRNA表達均明顯升高。 結論 1320nmND:YAG激光治療大鼠皮膚光老化效果顯著,其治療機制可能與其能夠刺激皮膚中Ⅰ型膠原、Ⅲ型膠原、TGF-β1mRNA表達升高有關。
[Abstract]:Background and objective skin photoaging mainly refers to the aging phenomenon of human skin affected by ultraviolet and other external environmental factors, mainly manifested in rough skin, wrinkle spots and so on, which not only affects a person's appearance. And there is a positive correlation with skin cancer, its pathogenesis is not fully clear. There are many treatments for skin photoaging. YAG laser therapy is a wrinkle tightening technique developed in recent years, which has the characteristics of small trauma and rapid clinical healing, and has achieved good results in clinical treatment of skin photoaging. However, the specific treatment mechanism is not clear at present. In this study, the mechanism of skin photoaging and YAG laser therapy was further studied through the animal model of skin photoaging in rats, in order to be helpful to the clinical treatment of skin photoaging.
