磷脂酰肌醇3激酶調節(jié)亞基2在卡波西肉瘤中的表達及意義

【摘要】:目的:明確卡波西肉瘤中磷脂酸肌醇3調節(jié)亞基2(PIK3R2)蛋白及mRNA的表達水平,并分析PIK3R2表達與卡波西肉瘤不同臨床分期及病理特征的關系。方法:選取維吾爾族卡波西肉瘤患者30例和健康者24例的皮損石蠟包埋組織,采用免疫組織化學和實時熒光定量聚合酶鏈式反應(qPCR)方法檢測PIK3R2的表達水平,分析與性別、年齡、病理分期、皮損面積、HIV感染等臨床病理特征間關系。結果:卡波西肉瘤組中PIK3R2的蛋白表達陽性率為60%,正常皮膚組中為25%,有統計學意義(P0.05)。在卡波西肉瘤組內斑片期陽性表達率33.3%,斑塊期陽性率71.4%,結節(jié)期陽性率81.8%,有統計學意義(P0.05),后兩組間無統計學差異(P0.05),其余兩兩比較均有統計學差異,病理分期與蛋白表達量呈正相關(P0.05),在性別、年齡、皮損面積、HIV及HHV-8感染情況等方面無差異(P0.05)?úㄎ魅饬鼋M中PIK3R2的mRNA表達水平(2.92±0.56)顯著高于正常皮膚組(1.13±0.82),差異有統計學意義(P0.05),PIK3R2 mRNA的表達量在卡波西肉瘤組織斑片期、斑塊期、結節(jié)期分別為2.37±0.20,2.83±0.54,3.56±0.23,三組之間差異均有統計學意義(P0.05),病理分期與m RNA表達量呈正相關(P0.05),但與卡波西肉瘤患者性別、年齡、皮損面積、HIV及HHV-8感染情況無關(P0.05)。結論:PIK3R2在卡波西肉瘤組織中的呈現高表達,PIK3R2的蛋白及m RNA表達水平在斑片期、斑塊期、結節(jié)期呈現遞增趨,呈正相關性,PIK3R2表達水平與患者的性別、年齡、皮損面積、HIV及HHV-8感染情況無關,PIK3R2可為尋找卡波西肉瘤的分子生物診斷標記提供一定基礎,為判斷卡波西肉瘤的病理演變提供新思路、新線索。
[Abstract]:Objective: to determine the expression of phosphatidylinositol 3 mediated subunit 2 (PIK3R2) protein and mRNA in the sarcoma of card Posey's sarcoma, and to analyze the relationship between PIK3R2 expression and the different clinical stages and pathological features of Posey sarcoma. Methods: 30 cases of Uygur card Posey sarcoma and 24 cases of healthy persons were selected to be embedded in paraffin embedded tissue, and immunohistochemistry was used. The expression level of PIK3R2 was detected by real time fluorescence quantitative polymerase chain reaction (qPCR), and the relationship between the sex, age, pathological stage, skin lesion area, HIV infection and other clinicopathological features was analyzed. Results: the positive rate of PIK3R2 protein expression in the group of card Posey's sarcoma was 60% and 25% in the normal skin group (P0.05). In card Posey The positive rate of patch stage in the sarcoma group was 33.3%, the positive rate of plaque stage was 71.4%, the positive rate of nodular stage was 81.8%, with statistical significance (P0.05). There was no statistical difference between the two groups (P0.05), the other 22 were statistically different, the pathological stage and the protein expression Cheng Zhengxiang (P0.05), in sex, age, skin lesion area, HIV and HHV-8 infection There was no difference in the other aspects (P0.05). The level of mRNA expression of PIK3R2 in the carPosey's sarcoma group was significantly higher than that of the normal skin group (1.13 + 0.82), the difference was statistically significant (P0.05). The expression of PIK3R2 mRNA was 2.37 + 0.20,2.83 + 0.54,3.56 + 0.23, respectively, in the tissue plaque stage of the sarcoma of the card Posey's sarcoma, respectively, and the three groups were different among the three groups. Statistical significance (P0.05), the pathological staging was positively correlated with the expression of M RNA (P0.05), but was not related to the sex, age, skin lesion area, HIV and HHV-8 infection in patients with card Posey's sarcoma (P0.05). Conclusion: PIK3R2 is highly expressed in the sarcoma of the card, and the expression level of PIK3R2 protein and m RNA is at the stage of patch, patch period, and nodular phase. There is a positive correlation. The level of PIK3R2 expression is not related to the sex, age, skin lesion area, HIV and HHV-8 infection. PIK3R2 can provide a certain basis for the molecular biomarkers for finding the card Posey's sarcoma, and provide new clues for judging the pathological changes of Capocci's sarcoma.
