【摘要】： 目的：探討真皮多能干細(xì)胞(dermal multipotential stem cells, DMSCs)對(duì)大鼠皮膚光老化(skin photoage)是否具有修復(fù)作用,為皮膚光老化的治療提供新的思路。方法：將48只SD大鼠隨機(jī)分組：正常組8只和光老化模型組40只,采用UVA和UVB紫外燈同時(shí)對(duì)模型組進(jìn)行照射(每天2h,連續(xù)照射60天)后,隨機(jī)取8只大鼠采用HE染色法進(jìn)行皮膚光老化模型鑒定,確定建模成功后隨機(jī)將模型組大鼠分為治療組16只和對(duì)照組16只。將體外分離培養(yǎng)鑒定的SD乳鼠DMSCs (1×106細(xì)胞懸液)多點(diǎn)注射于治療組大鼠光老化區(qū)皮膚內(nèi),每?jī)芍芤淮?而對(duì)照組用PBS替代處理。分別對(duì)正常組、對(duì)照組、治療組第4周、治療組第8周大鼠皮膚進(jìn)行取材,采用免疫組織化學(xué)染色法檢測(cè)大鼠表皮中P53和PCNA的表達(dá)。結(jié)果：經(jīng)免疫組織化學(xué)染色法檢測(cè)顯示：與對(duì)照組相比,4周治療組大鼠表皮中的P53陽(yáng)性表達(dá)無明顯變化,兩者比較其差異無統(tǒng)計(jì)學(xué)意義(P0.05)；8周治療組大鼠表皮中的P53陽(yáng)性表達(dá)減弱,兩者比較其差異有統(tǒng)計(jì)學(xué)意義(P0.05)；4周和8周治療組大鼠表皮中的PCNA的陽(yáng)性表達(dá)均有增強(qiáng),治療前后比較均有統(tǒng)計(jì)學(xué)意義(P0.05)。8周治療組P53和PCNA與正常對(duì)照組比較均無統(tǒng)計(jì)學(xué)差異性(P0.05)。結(jié)論：DMSCs大鼠皮膚內(nèi)注射后可以抑制光老化皮膚P53蛋白表達(dá)、增強(qiáng)PCNA的表達(dá),兩者的表達(dá)與其在正常大鼠表皮中的表達(dá)相一致,考慮DMSCs對(duì)光老化可能有一定的修復(fù)作用。
[Abstract]:Objective: To investigate the effects of dermal multipotent stem cells (dermal multipotential stem cells, DMSCs) on rat skin photoaging (skin photoage) with repair effect, to provide new ideas for the treatment of skin photoaging. Methods: 48 SD rats were randomly divided into normal group of 8 rats and aging model group with 40 rats. Using UVA and UVB UV light irradiation on the model group at the same time (2H per day, continuous 60 days after irradiation), 8 rats were randomly selected using HE skin photoaging model identification staining identified were established and the model rats were divided into treatment group 16 and control group 16 rats. In vitro isolation and identification of SD rat DMSCs (1 * 106 cell suspension) multi point injection in the treatment of rats photoaging skin, once every two weeks, while the control group treated with PBS respectively.
