糖尿病大鼠肝臟組織中維生素D受體表達(dá)與血清25-羥維生素D相關(guān)性研究

【摘要】:目的檢測(cè)T2DM大鼠肝臟中維生素D受體(VDR)mRNA及蛋白表達(dá),探討T2DM大鼠肝臟VDR表達(dá)及血清25-羥維生素D[25(OH)D]與代謝指標(biāo)的相關(guān)性。方法取60只健康雄性Wistar大鼠,隨機(jī)分成T2DM組(n=30)和正常對(duì)照(NC)組(n=30)。T2DM組予高脂高膽固醇飼料喂養(yǎng)4周后,禁食16 h,按30 mg/kg單次腹腔注射STZ,72 h后尾靜脈采血測(cè)血糖,以血糖濃度16.7mmol/L作為糖尿病模型造模成功的標(biāo)準(zhǔn)。ELISA測(cè)定血清25(OH)D水平,RT-PCR和Western blot分別檢測(cè)兩組肝臟組織中VDR mRNA及蛋白表達(dá)。Spearman相關(guān)分析T2DM組血清25(OH)D與FPG、F1ns、FFA、TG、TC、HOMA-IR的關(guān)系。結(jié)果與NC組比較,T2DM組肝臟中VDR mRNA及蛋白表達(dá)增高[(2.15±0.25)vs(8.26±2.23),(0.317±0.085)vs(0.542±0.135)],血清25(OH)D水平降低[(43.6±12.3)vs(21.3±6.7)ng/ml],血清FFA、TG、TC、LDL-C、FIns、HOMA-IR增高(P0.05)。相關(guān)性分析顯示,血清25(OH)D與FFA、TG、FIns、HOMA-IR呈負(fù)相關(guān),與HDL-C呈正相關(guān)(P0.05)。結(jié)論 T2DM大鼠肝臟組織中VDR mRNA及蛋白表達(dá)增加,而血清25(OH)D降低。血清25(OH)D降低可能會(huì)反饋性促進(jìn)肝臟VDR表達(dá),并可能進(jìn)一步導(dǎo)致肝臟IR及糖脂代謝異常。
[Abstract]:Aim to detect the expression of vitamin D receptor (VDR) (VDR) mRNA and protein in the liver of T2DM rats, and to explore the relationship between the expression of VDR and serum 25-hydroxyvitamin D [25 (OH) D] in the liver of T2DM rats. Methods Sixty healthy male Wistar rats were randomly divided into two groups: T2DM group (n = 30) and normal control group (n = 30). T2DM group was fed with high fat and high cholesterol diet for 4 weeks and fasting for 16 hours. STZ, was injected intraperitoneally at 30 mg/kg for 16 hours. 72 hours later, blood samples were collected from tail vein to measure blood glucose. Blood glucose concentration (16.7mmol/L) was used as the standard for successful establishment of diabetic model. Serum 25 (OH) D levels were measured by Elisa. The expression of VDR mRNA and protein in liver tissue of two groups were detected by RT-PCR and Western blot respectively. Spearman correlation analysis was used to analyze the relationship between serum 25 (OH) D and FPG,F1ns,FFA,TG,TC,HOMA-IR in T2DM group. Results compared with NC group, the expression of VDR mRNA and protein in liver of T2DM group was increased [(2.15 鹵0.25) vs (8.26 鹵2.23), (0.317 鹵0.085) vs (0.542 鹵0.135)]. Serum 25 (OH) D level decreased [(43.6 鹵12.3) vs (21.3 鹵6.7) ng/ml] and serum FFA,TG,TC,LDL-C,FIns,HOMA-IR increased (P0.05). Correlation analysis showed that serum 25 (OH) D was negatively correlated with FFA,TG,FIns,HOMA-IR and positively correlated with HDL-C (P0.05). Conclusion the expression of VDR mRNA and protein in liver tissue of T2DM rats increased, while serum 25 (OH) D decreased. The decrease of serum 25 (OH) D may promote the expression of VDR in liver and further lead to abnormal metabolism of IR and glucose and lipid in liver.
