【摘要】：目的探討內(nèi)質(zhì)網(wǎng)應(yīng)激信號(hào)通路在高脂誘導(dǎo)的心肌細(xì)胞損傷中的作用。方法棕櫚酸(0.1~0.4 mmol·L~(-1))刺激心肌細(xì)胞(0~48 h),CCK-8法評(píng)估細(xì)胞生長(zhǎng)狀態(tài),免疫印跡法評(píng)估細(xì)胞內(nèi)內(nèi)質(zhì)網(wǎng)應(yīng)激信號(hào)通路相關(guān)蛋白表達(dá)水平及細(xì)胞凋亡蛋白表達(dá)水平。結(jié)果棕櫚酸(0.1~0.4 mmol·L~(-1))刺激H9C2心肌細(xì)胞24 h,0.2、0.4 mmol·L~(-1)組細(xì)胞增殖率均出現(xiàn)明顯下降。棕櫚酸(0.2 mmol·L~(-1))刺激H9C2心肌細(xì)胞時(shí),24、48 h組細(xì)胞增殖率均出現(xiàn)明顯下降。棕櫚酸(0.2 mmol·L~(-1))刺激24 h,GRP78、CHOP、PERKphos、IRE1phos、ATF6等內(nèi)質(zhì)網(wǎng)應(yīng)激相關(guān)蛋白及Bax蛋白表達(dá)均明顯增高(P0.05),而B(niǎo)cl-2表達(dá)出現(xiàn)明顯降低(P0.05)。預(yù)處理內(nèi)質(zhì)網(wǎng)應(yīng)激抑制劑普伐他汀(pravastatin,10mol·L~(-1))的棕櫚酸(0.2 mmol·L~(-1))組與不加pravastatin的棕櫚酸(0.2 mmol·L~(-1))組相比,Bcl-2及Bax的表達(dá)均恢復(fù)至正常水平。結(jié)論內(nèi)質(zhì)網(wǎng)應(yīng)激信號(hào)通路對(duì)高脂引起的心肌損傷的發(fā)病及治療均起到重要作用。
[Abstract]:Objective to investigate the role of endoplasmic reticulum stress signaling pathway in hyperlipidemic cardiomyocyte injury. Methods the growth status of cardiomyocytes was evaluated by palmitic acid (0.1 ~ 0.4 mmol L ~ (-1) (0 ~ 48 h), CCK-8), and the expression level of intracellular endoplasmic reticulum stress signal pathway associated protein and apoptotic protein were evaluated by Western blot. Results the proliferation rate of H9C2 cardiomyocytes stimulated by palmitic acid (0.1 ~ 0.4 mmol / L ~ (-1) at 24 h or 0.2 mmol L ~ (-1) was significantly decreased. When palmitic acid (0.2 mmol L ~ (-1) stimulated H9C2 cardiomyocytes, the proliferation rate of H9C2 cells decreased significantly in 24 h group. Palmitic acid (0.2 mmol L ~ (-1) increased the expression of ER stress-related protein and Bax protein (P0.05), while the expression of Bcl-2 decreased significantly (P0.05). The expression of Bcl-2 and Bax returned to normal level in pravastatin,10mol L ~ (-1) preconditioning group compared with pravastatin free palmitic acid (0.2 mmol L ~ (-1) group). Conclusion endoplasmic reticulum stress signaling pathway plays an important role in the pathogenesis and treatment of hyperlipidemic myocardial injury.
