【摘要】：目的:探究胰島素受體底物2(insulin receptor substrate 2,IRS2)基因單核苷酸多態(tài)性位點Gly1057Asp與環(huán)境間的交互作用對2型糖尿病(type 2 diabetes mellitus,T2DM)易感性的影響。方法:對772例無血緣關系的中國重慶漢族人(T2DM 467例,健康對照305例)進行病例-對照研究。應用限制性片段長度多態(tài)性聚合酶鏈反應聯(lián)合基因測序法檢測IRS2基因Gly1057Asp位點基因型,分析其與T2DM的相關性。采用多因子降維法(multifactor dimensionality reduction,MDR)分析Gly1057Asp位點與老齡、肥胖及高甘油三酯血癥間的交互作用對T2DM易感性的影響。結(jié)果:IRS2基因Gly1057Asp位點與T2DM相關,GG基因型個體T2DM患病風險是AA基因型的1.42倍(OR=1.422,95%CI=1.037~1.949,P=0.029)。MDR分析顯示Gly1057Asp位點、年齡、腰圍和血漿三酰甘油四個因素構(gòu)成的交互模型具有統(tǒng)計學意義(P=0.001)。在老齡(年齡60歲)或腹型肥胖者中,Gly1057Asp位點GG或GA基因型個體T2DM患病風險明顯增加;在腹型肥胖的高甘油三酯血癥者中,除年齡在40~60歲且具AA基因型者外,其余均易患T2DM(OR=3.342,95%CI=1.168~9.561,P=0.026)。結(jié)論:同時具有IRS2基因Gly1057Asp位點GG基因型、腹型肥胖和高甘油三酯血癥的中國重慶漢族人,可能是T2DM的高危易感人群。
[Abstract]:Objective: To investigate the effect of the interaction between the single nucleotide polymorphism site (Gly1057Asp) of the insulin receptor substrate 2 (insulin receptor substrate 2, IRS2) gene and the environment on type 2 diabetes mellitus (type 2 diabetes mellitus, T2DM) susceptibility. Methods: 772 cases of Chongqing Han people in China (T2DM 467 cases, 305 healthy controls) were carried out. Case control study. The restriction fragment length polymorphism polymerase chain reaction (PCR) combined gene sequencing was used to detect the genotype of IRS2 gene Gly1057Asp, and the correlation with T2DM was analyzed. The multiple factor dimensionality reduction (multifactor dimensionality reduction, MDR) was used to analyze the Gly1057Asp locus with aging, obesity and hypertriglyceridemia. The effect of interaction on T2DM susceptibility. Results: the Gly1057Asp locus of the IRS2 gene is associated with T2DM. The risk of T2DM disease in the GG genotype is 1.42 times the AA genotype (OR=1.422,95%CI=1.037~1.949, P=0.029).MDR analysis shows the Gly1057Asp site, age, waist circumference and plasma three glycerol four factors have statistical meaning. Meaning (P=0.001). Among the elderly (age 60) or abdominal obesity, the risk of T2DM in the Gly1057Asp locus GG or GA genotype individuals increased significantly; among those with high triglyceride in abdominal obesity, the rest were susceptible to T2DM (OR= 3.342,95%CI=1.168~9.561, P=0.026) except for the age of 40~60 and the AA genotypes. Gly1057Asp locus GG genotype, abdominal obesity and hypertriglyceridemia in Han Chinese in Chongqing, China, may be a high-risk susceptible group of T2DM.
