不同濃度異丙酚對哮喘大鼠氧化應(yīng)激反應(yīng)及氣道炎癥的影響

【摘要】:目的探討不同濃度異丙酚對哮喘大鼠氧化應(yīng)激反應(yīng)及氣道炎癥的影響。方法清潔級SD大鼠32只隨機(jī)分為正常組、模型組、低劑量組、高劑量組,每組大鼠8只。正常組給予等量生理鹽水灌胃;模型組給予等量生理鹽水灌胃,且于22 d霧化吸入后腹腔注射50 mg/kg戊巴比妥鈉麻醉;低劑量組、高劑量組分別給予25 mg·kg~(-1)·h~(-1)和50 mg·kg~(-1)·h~(-1),靜脈滴注。各組大鼠均在治療后7 d取器官組織行HE染色觀察。比較各組大鼠血清白介素(IL)-5、分離嗜酸性粒細(xì)胞(EOS)、免疫球蛋白(Ig)E、IL-4水平及肺組織勻漿超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)水平變化。結(jié)果與正常組比較,模型組、低劑量組、高劑量組IL-5、EOS、Ig E和IL-4水平明顯增加,差異有統(tǒng)計學(xué)意義(P0.05);低劑量組和高劑量組IL-5、EOS、Ig E和IL-4水平明顯低于模型組(P0.05);高劑量組IL-5、EOS、Ig E和IL-4水平明顯低于低劑量組(P0.05);與正常組比較,模型組、低劑量組、高劑量組MDA水平明顯增加,而SOD水平明顯降低(P0.05);低劑量組和高劑量組MDA水平明顯低于模型組,而SOD水平明顯高于模型組(P0.05);高劑量組MDA水平明顯低于低劑量組,而SOD水平明顯高于低劑量組(P0.05)。結(jié)論不同濃度異丙酚可抑制哮喘模型大鼠增高的氧化應(yīng)激反應(yīng),以及緩解大鼠氣道炎癥,其中以50 mg·kg~(-1)·h~(-1)濃度的異丙酚效果更為明顯。
[Abstract]:Objective to investigate the effects of propofol at different concentrations on oxidative stress and airway inflammation in asthmatic rats. Methods Thirty-two clean SD rats were randomly divided into normal group, model group, low dose group and high dose group, with 8 rats in each group. The normal group was given the same amount of normal saline intragastrically, the model group was given the same amount of normal saline, and 50 mg/kg pentobarbital sodium was injected intraperitoneally after 22 days of atomization inhalation. All the rats in each group were observed by HE staining on the 7th day after treatment. Serum IL-5, eosinophil, immunoglobulin IgE, IL-4 and the levels of superoxide dismutase (SOD), malondialdehyde (MDA) MDAA, nitric oxide (no) in lung homogenate were compared in each group. Results compared with the normal group, the levels of IL-5 EOS E and IL-4 increased significantly in the model group, low dose group and high dose group. The levels of Ig E and IL-4 in low dose group and high dose group were significantly lower than those in model group, and the levels of IL-5 EOSG E and IL-4 in high dose group were significantly lower than those in low dose group (P 0.05), and in model group, low dose group, compared with normal group, the level of Ig E and IL-4 in high dose group was significantly lower than that in low dose group. The level of MDA in the high dose group was significantly increased, while the level of SOD was significantly decreased, the MDA level in the low dose group and the high dose group was significantly lower than that in the model group, while the SOD level in the high dose group was significantly higher than that in the model group, and the MDA level in the high dose group was significantly lower than that in the low dose group. The level of SOD was significantly higher than that of low dose group (P 0.05). Conclusion different concentrations of propofol can inhibit the increased oxidative stress response and relieve airway inflammation in asthmatic rats. The effects of propofol at the concentration of 50 mg 路kg ~ (-1) 路h ~ (-1) 路h ~ (-1) are more obvious.
