二苯乙烯苷對(duì)糖尿病大鼠心肌損傷的保護(hù)作用

【摘要】:目的探討二苯乙烯苷(TSG)對(duì)糖尿病大鼠心肌損傷的作用及對(duì)沉默信息調(diào)節(jié)因子2的哺乳動(dòng)物同源體1(SIRT1)和磷酸腺苷活化的蛋白激酶(AMPK)蛋白影響。方法建立2型糖尿病大鼠模型,分組給藥,16周時(shí)處死大鼠,生化法測(cè)定血糖、血脂、肝功能及肌酸激酶(CK)、乳酸脫氫酶(LDH)和心肌組織游離脂肪酸(NEFA);酶聯(lián)免疫吸附法測(cè)定心肌肌鈣蛋白Ⅰ(cTnⅠ)、心肌組織脂代謝相關(guān)酶脂肪酸跨膜轉(zhuǎn)運(yùn)載體蛋白(FATPs)和脂肪酸β氧化酶(FA-β-oxidase)的含量;放射免疫法測(cè)定血漿炎癥因子腫瘤壞死因子(TNF-α)、白細(xì)胞介素6(IL-6)、白細(xì)胞介素1β(IL-1β)的含量,Western blot檢測(cè)心肌組織中TNF-α、IL-6、IL-1β、SIRT1和AMPK蛋白的表達(dá);并測(cè)定左心室膠原含量。結(jié)果TSG干預(yù)后能減低血脂水平,對(duì)血糖和胰島素水平無(wú)明顯影響。TSG能減低糖尿病大鼠心肌組織中膠原含量,減少心臟游離脂肪酸含量,增加心肌組織脂代謝相關(guān)酶(FATPs和FA-β-oxidase)含量,抑制外周血和心肌組織中炎癥因子(TNF-α、IL-6、IL-1β)的分泌。TSG能明顯增加糖尿病大鼠心臟SIRT1和p AMPK蛋白表達(dá)。結(jié)論 TSG對(duì)糖尿病大鼠心肌具有保護(hù)作用,其機(jī)制可能與抑制心肌炎癥因子和改善能量代謝有關(guān)。
[Abstract]:Objective to investigate the effects of stilbene glucoside (TSGG) on myocardial injury in diabetic rats and its effects on mammalian homologue 1 SIRT1 (silencing signaling factor 2) and adenosine phospho-activated protein kinase (AMPK) protein.
