對講機(jī)使用產(chǎn)生的電磁暴露對Wif-1基因甲基化水平及血液中淋巴細(xì)胞構(gòu)成的影響

【摘要】:有研究提示電磁暴露可能是白血病發(fā)病的潛在危險(xiǎn)因素。為探討對講機(jī)產(chǎn)生的電磁暴露對Wif-1基因甲基化水平及血液中白細(xì)胞構(gòu)成的影響,采用1:1配對設(shè)計(jì),選擇年齡匹配的60名男性作為研究對象,最終納入分析的為對講機(jī)使用組(暴露組)29名,非使用組(對照組)28名。采用自行設(shè)計(jì)的流行病學(xué)調(diào)查問卷收集研究對象的一般人口學(xué)特征及職業(yè)暴露情況等。采用美國SEQUENOM Mass Array?Epi TYPER技術(shù)平臺(tái)檢測Wif-1基因的甲基化水平。采用配對t檢驗(yàn)和多因素混合效應(yīng)模型分析對講機(jī)使用和Wif-1基因甲基化水平,以及Wif-1基因甲基化水平和血液中白細(xì)胞構(gòu)成的關(guān)系。研究結(jié)果顯示:在Wif-1基因啟動(dòng)子區(qū)域的32個(gè)CpG位點(diǎn)中,7個(gè)位點(diǎn)(CpG7、CpG8、CpG12、CpG16、CpG21、CpG28和CpG31)的甲基化水平暴露組顯著高于對照組;電磁暴露與Wif-1基因平均甲基化水平呈正相關(guān)(偏回歸系數(shù)β=2.19,概率P=0.026)。而Wif-1基因甲基化水平與淋巴細(xì)胞百分比呈正相關(guān)(β=0.633,P=0.020),與嗜酸性粒細(xì)胞百分比呈負(fù)相關(guān)(β=-0.151,P=0.016)。結(jié)果表明電磁暴露可能通過調(diào)節(jié)Wif-1基因甲基化水平而影響血液中淋巴細(xì)胞的構(gòu)成,因而遺傳的改變可能是電磁暴露致癌的重要機(jī)制,但是還需要更大樣本量的研究來進(jìn)一步確認(rèn)。
[Abstract]:Some studies have suggested that electromagnetic exposure may be a potential risk factor for leukemia. In order to investigate the effect of electromagnetic exposure on the methylation level of Wif-1 gene and the leukocyte composition in blood, a 1: 1 pairing design was used. Sixty age-matched men were selected as study subjects, and the interphone use group (29 exposed group) was included in the analysis. A self-designed epidemiological questionnaire was used to collect the general demographic characteristics and occupational exposure of the subjects in the non-use group (control group, n = 28). The methylation level of Wif-1 gene was detected by Epi TYPER technique platform. The level of interphone use and Wif-1 gene methylation was analyzed by paired t test and multivariate mixed effect model. The relationship between the methylation level of Wif-1 gene and leukocyte composition in blood. The results showed that the methylation level of CpG7, CpG8, CpG12, CpG21, CpG21 and CpG31 in the 32 CpG loci of the promoter region of the Wif-1 gene were significantly higher in the exposed group than in the control group. There was a positive correlation between electromagnetic exposure and average methylation level of Wif-1 gene (partial regression coefficient 尾 2.19, probabilistic P = 0.026%), but a positive correlation between Wif-1 gene methylation level and lymphocyte percentage (尾 -0.633G, P < 0.020), and a negative correlation between Wif-1 gene methylation level and percentage of eosinophil (尾 -0.151). The results showed that the methylation level of Wif-1 gene was positively correlated with the percentage of lymphocytes. Electromagnetic exposure may affect the composition of lymphocytes in the blood by regulating the methylation level of the Wif-1 gene. Genetic changes may be an important mechanism of carcinogenesis in electromagnetic exposure, but more studies are needed to further confirm it.
