外源性心鈉素對小鼠過敏性哮喘氣道炎癥的影響及其機制分析

【摘要】:目的探討外源性心鈉素對過敏性哮喘小鼠模型氣道炎癥的影響作用及其作用機制。方法選取SPF級BALBc小鼠48只,采用隨機數字表法分為對照組(等量生理鹽水干預)、模型組(等量生理鹽水干預)、實驗組A(外源性心鈉素0.5μg/g)、實驗組B(外源性心鈉素0.5μg/g+A71915)。模型組、實驗組A、實驗組B建模成功后給予對應處理措施,在最后一次激發(fā)實驗后24h,采集各組血清、支氣管肺泡灌洗液檢查炎癥因子,采用HE染色觀察肺組織病理變化,采用Western blot技術檢測肺組織中鋅指轉錄因子(GATA3)蛋白的表達水平。結果模型組小鼠的血液中EOS、淋巴細胞、中性粒細胞所占比例均高于對照組(P0.05),實驗組A的血液中EOS、淋巴細胞、中性粒細胞所占比例均高于模型組、實驗組B(P0.05);模型組小鼠的肺泡灌洗液中EOS、淋巴細胞、中性粒細胞計數、IL-6、TNF-α水平均高于對照組(P0.05),實驗組A的肺泡灌洗液中EOS、淋巴細胞、中性粒細胞計數、IL-6、TNF-α水平均高于模型組、實驗組B(P0.05);模型組小鼠的肺組織中GATA3蛋白表達水平均高于對照組(P0.05),實驗組A肺組織中GATA3蛋白表達水平高于模型組、實驗組B(P0.05)。結論外源性心鈉素會進一步加劇過敏性哮喘小鼠模型的氣道炎癥反應,與激活炎性細胞、炎癥因子釋放及增強肺組織中GATA3蛋白表達有關。
[Abstract]:Objective to investigate the effect and mechanism of exogenous atrial natriuretic peptide (ANP) on airway inflammation in allergic asthmatic mice. Methods 48 SPF grade BALBc mice were selected. The rats were randomly divided into control group, model group, experimental group A (external atrial natriuretic peptide 0.5 渭 g / g) and experimental group B (external atrial natriuretic peptide 0.5 渭 g / g A71915). Experimental group A and group B were given corresponding treatment after successful modeling. 24 hours after the last exciting experiment, the serum and bronchoalveolar lavage fluid of each group were collected to examine the inflammatory factors, and the pathological changes of lung tissue were observed by HE staining. Western blot technique was used to detect the expression of zinc finger transcription factor (GATA3) protein in lung tissue. Results the percentage of EOS, lymphocytes and neutrophils in the blood of the model group was higher than that in the control group (P 0.05), and that in the blood of the experimental group A was higher than that of the control group. The percentage of neutrophils in the model group was higher than that in the model group, and the levels of EOS, lymphocytes and neutrophils in the alveolar lavage fluid of the model group were higher than those in the control group, and the levels of EOS and lymphocytes in the alveolar lavage fluid of the experimental group were higher than those in the control group. The level of IL-6 TNF- 偽 in the model group was higher than that in the model group, and the expression level of GATA3 protein in the lung tissue of the model group was higher than that of the control group (P 0.05), and the expression level of GATA3 protein in the lung tissue of the experimental group was higher than that of the model group. Conclusion exogenous atrial natriuretic peptide can further aggravate the airway inflammation in allergic asthma mice, which is related to the activation of inflammatory cells, the release of inflammatory factors and the enhancement of GATA3 protein expression in lung tissue.
