高糖高溶血磷脂膽堿對系膜細胞產生纖維連接蛋白、Ⅳ型膠原及轉化生長因子-β1的影響

【摘要】:目的探討高糖高溶血磷脂膽堿環(huán)境對人腎小球系膜細胞(HMCs)產生纖維連接蛋白(Fn)、Ⅳ型膠原(ColⅣ)及轉化生長因子-β1(TGF-β1)的影響。方法將HMCs分為對照組、TGF-β1中和性抗體組、高糖高脂組、高糖高脂+TGF-β1中和性抗體組,采用qRT-PCR檢測TGF-β1mRNA的表達,ELISA檢測細胞上清液中ColⅣ及Fn含量。結果 (1)高糖高脂組TGF-β1mRNA表達水平較對照組升高(P0.05);(2)高糖高脂組細胞上清液TGF-β1、ColⅣ和Fn含量較對照組增加(P0.05),而這種作用可被TGF-β1中和性抗體所抑制(P0.05)。結論高糖高脂可通過促進系膜細胞TGF-β1表達從而誘導ECM的分泌,增加腎纖維化發(fā)生的風險。
[Abstract]:Objective to investigate the effects of high glucose and high lysophosphatidylcholine (LPC) environment on the production of fibronectin (FN) and transforming growth factor- 尾 1 (TGF- 尾 1) in human glomerular Mesangial cells (HMCs). Methods HMCs was divided into two groups: the control group with TGF- 尾 1 neutralizing antibody, the group with high glucose and high fat, the control group with TGF- 尾 1 neutralizing antibody and the group with high glucose and high fat. High glucose and high fat TGF- 尾 1 neutralizing antibody group, The expression of TGF- 尾 _ 1 mRNA in supernatant was detected by qRT-PCR and Elisa.
