酪酸梭菌對食物過敏小鼠腸道屏障功能的影響

【摘要】:目的探討產丁酸的益生菌酪酸梭菌通過調節(jié)腸上皮細胞雙孔鉀通道Trek1的表達對過敏小鼠腸道屏障功能的影響。方法建立小鼠食物過敏模型,ELISA法、流式細胞儀檢測相關指標驗證造模效果,尤斯室檢測小腸組織通透性變化,Western blot及免疫熒光法檢測Na6ve組、Saline組、SIT組、SIT/SB組、SB組、SIT/CB組、CB組、SIT/CB/Spadin組小鼠空腸組織中Trek1的表達;Transwell系統(tǒng)中使用T84細胞建立單層上皮細胞層,分別暴露于過敏介質,qRT-PCR和Western blot檢測Trek1mRNA和蛋白的表達;分別使用生理鹽水、SIT、SIT/SB、SB、SIT/CB、CB、SIT/CB/Spadin不同療法對過敏小鼠進行治療,了解小鼠腸道Trek1的表達情況、腸道屏障功能及過敏反應指標的變化情況。結果相對于對照組,食物過敏小鼠小腸Trek1蛋白表達水平顯著下降,腸黏膜通透性顯著升高,差異有統(tǒng)計學意義(P0.05);T84細胞暴露于過敏介質后,Trek1mRNA和蛋白的表達顯著下降(P0.05),但預先加入p38抑制劑可以拮抗這種改變;SIT治療、酪酸梭菌和丁酸鈉單獨應用均可以提高食物過敏小鼠腸道Trek1的表達,減輕過敏反應(P0.05),但SIT與酪酸梭菌或丁酸鈉的聯合應用效果更加顯著(與SIT組相比P0.05),并且可以顯著降低小腸黏膜通透性,改善腸道屏障功能(P0.05)。結論丁酸鈉或者產丁酸的益生菌酪酸梭菌可以通過提高Trek1的表達來恢復過敏小鼠的腸道屏障功能,減輕過敏反應,并且加強SIT治療的效果。
[Abstract]:Objective to investigate the effects of Clostridium caseinate, a probiotics producing butyric acid, on intestinal barrier function in allergic mice by regulating the expression of double-pore potassium channel Trek1 in intestinal epithelial cells. ELISA and flow cytometry were used to test the effect of model making. The changes of intestinal permeability were detected by Eus chamber. The changes of intestinal permeability were detected by Western blot and immunofluorescence were used to detect the Na6ve group. The expression of Trek1 in jejunum of Saline / sit / sit / SB / SB / CB / CB / CB group. T84 cells were used in the Transwell system to establish a single layer of epithelial cells, which were exposed to allergy mediators respectively. QRT-PCR and Western blot were used to detect the expression of Trek1mRNA and protein. The allergic mice were treated with different treatments of normal saline SITS / SBT / SBT / CBT / CBST / CB/ Spadin. Results compared with the control group, the expression of Trek1 protein in the small intestine of mice with food allergy was significantly lower than that in the control group. The permeability of intestinal mucosa was significantly increased (P 0.05). The mRNA and protein expression of Trek1 in T84 cells decreased significantly after exposure to allergy mediators, but p38 inhibitor could antagonize the changes. SIT treatment, Clostridium caseinate and sodium butyrate alone can increase the expression of Trek1 in the intestine of food allergy mice and alleviate the allergic reaction (P0.05). However, the combination of SIT with Clostridium caseinate or sodium butyrate was more effective (compared with SIT group, P 0.05), and could significantly reduce intestinal mucosal permeability. Conclusion Sodium butyrate or butyric acid-producing probiotics Clostridium caseate can restore the intestinal barrier function of allergic mice by increasing the expression of Trek1 and alleviate the allergic reaction. And enhance the effect of SIT treatment.
