短期PM 2.5 暴露所致腎臟損傷的探討及復(fù)方精油預(yù)防作用的研究

目的:隨著空氣污染的加劇,PM2.5近年來(lái)倍受關(guān)注,然而研究大多局限于PM2.5對(duì)呼吸系統(tǒng)的影響,對(duì)腎臟的影響研究較少。本實(shí)驗(yàn)探討了在PM2.5短期暴露造成急性肺損傷的情況下腎臟的損傷情況。同時(shí),探究了復(fù)方精油是否可對(duì)這種損傷進(jìn)行干預(yù)從而達(dá)到預(yù)防性治療的目的。方法:1.PM2.5樣品采集及PM2.5懸液制備:本次樣本采集于2013年中國(guó)河北廊坊,用專門的PM2.5采樣器于冬季收集。將收集完成的PM2.5石英濾膜放入超純水中浸泡、超聲處理2-3小時(shí),使PM2.5顆粒與濾膜分離懸浮于超純水中,后經(jīng)冷凍干燥去除水分。稱取一定量的PM2.5顆粒物,用無(wú)菌生理鹽水配置成10 mg/m L PM2.5懸液,使用前吹打混勻。2.實(shí)驗(yàn)動(dòng)物處理及實(shí)驗(yàn)設(shè)計(jì):6-8周的雄性Balb/c小鼠置于適宜的溫度(24±1oC),充足的水和食物,規(guī)律的晝夜循環(huán)環(huán)境中。本次的實(shí)驗(yàn)分為兩部分:(1)第一部分,48只BALB/c小鼠,隨機(jī)分成兩組(n=24),分別為空白對(duì)照組和PM2.5暴露實(shí)驗(yàn)組。對(duì)照組和PM2.5暴露組小鼠分別在實(shí)驗(yàn)的第0、2天通過(guò)氣管灌注方式滴注PM2.5懸液50μL。每次分別在對(duì)照組和PM2.5暴露...
