伊馬替尼對(duì)尿毒癥心肌病大鼠心肌纖維化的保護(hù)作用

【摘要】:目的觀察伊馬替尼通過(guò)調(diào)控PDGFRα的表達(dá)改善尿毒癥大鼠心肌纖維化的作用。方法將72只大鼠分為Sham組,5/6組和5/6+I組,5/6組建立大鼠尿毒癥模型,5/6+I組建模后行伊馬替尼灌胃,Sham組僅行腎臟游離。所有大鼠于術(shù)后8周行心臟病理染色;實(shí)時(shí)定量熒光PCR、免疫組織化學(xué)測(cè)定心臟血小板衍生生長(zhǎng)因子受體α(PDGFRα)mRNA及蛋白表達(dá)。結(jié)果 5/6組和5/6+I組大鼠尿蛋白、Scr、BUN均較Sham組明顯增加(P0.01).Sham組心肌病理評(píng)分顯著低于5/6組(P0.01),而5/6+I組顯著低于5/6組(P0.01);5/6組心肌間質(zhì)膠原容積分?jǐn)?shù)值明顯高于Sham組(P0.01);5/6+I組高于Sham組(P0.05),但與5/6組比較,明顯下降(P0.05);5/6組和5/6+I組的PDGFRαmRNA和蛋白表達(dá)均顯著高于Sham組(均P0.01);而5/6+I組明顯低于5/6組(P0.05)。結(jié)論伊馬替尼可以通過(guò)抑制PDGFRα表達(dá)減少尿毒癥大鼠心臟病理?yè)p傷及纖維化的程度。
[Abstract]:Aim to observe the effect of imatinib on myocardial fibrosis in uremic rats by regulating the expression of PDGFR 偽. Methods 72 rats were divided into Sham group, 5 / 6 group and 5 / 6 I group. The expression of platelet derived growth factor receptor 偽 (PDGFR 偽) mRNA and protein was detected by real-time quantitative PCR, immunohistochemistry. Results urinary protein and Scr,BUN of rats in 5 / 6 and 5 / 6 I groups were significantly higher than those in Sham group (P0.01). Sham group was significantly lower than that of 5 / 6 group (P0.01), and 5 / 6 I group was significantly lower than 5 / 6 group (P0.01); 5 / 6 group was significantly higher than Sham group (P0.01); 5 / 6 I group was significantly higher than that of Sham group (P0.01); group 5 / 6 I group was significantly lower than that of Sham group (P0.01); group 5 / 6 I group was significantly lower than 5 / 6 group (P0.01). Higher than Sham group (P0.05), but compared with 5 / 6 group, The expression of PDGFR 偽 mRNA and protein in 5 / 6 group and 5 / 6 I group was significantly higher than that in Sham group (P0.01), while the expression of PDGFR 偽 mRNA and protein in 5 / 6 I group was significantly lower than that in 5 / 6 group (P0.05). Conclusion imatinib can reduce the degree of cardiac injury and fibrosis in uremic rats by inhibiting the expression of PDGFR 偽.
