三氧化二砷對腎母細胞瘤增殖及凋亡的影響

【摘要】:目的觀察三氧化二砷(As2O3)對體外人腎母細胞瘤SK-NEP-1細胞株增殖和凋亡的影響。方法應用0、1.25、2.5、5、10μmol/L As2O3(A、B、C、D、E組)作用SK-NEP-1細胞24 h。采用MTT比色法檢測不同濃度As2O3對SK-NEP-1細胞的增殖抑制率;流式細胞儀檢測凋亡率;ELISA法檢測SK-NEP-1細胞Bcl-2水平變化;Caspase 3試劑盒檢測細胞Caspase 3活性。結果 A、B、C、D、E組細胞增殖抑制率分別為(3.25±1.05)%、(11.19±2.27)%、(20.72±2.21)%、(27.24±0.57)%、(32.25±2.15)%,細胞凋亡率分別為(5.12±0.29)%、(13.42±1.03)%、(23.70±0.97)%、(32.44±0.82)%、(40.22±1.31)%,各組間差異均具有統(tǒng)計學意義(均P0.05)。B、C、D、E組Bcl-2蛋白表達水平均低于A組(均P0.05),Caspase 3活性均高于A組(均P0.05)。結論 As2O3能明顯抑制腎母細胞瘤細胞的增殖,并進一步誘導其凋亡,其抗腫瘤機制可能與下調Bcl-2表達及激活Caspase 3有關。
[Abstract]:Objective to observe the effect of arsenic trioxide (as _ 2O _ 3) on proliferation and apoptosis of human nephroblastoma cell line SK-NEP-1 in vitro. Methods SK-NEP-1 cells were treated with 10 渭 mol / L as _ 2O _ 3 10 渭 mol 路L ~ (2 +) C _ 2O _ 3 for 24 h. MTT assay was used to detect the proliferation inhibition rate of SK-NEP-1 cells at different concentrations of as _ 2O _ 3, and flow cytometry (FCM) was used to detect the apoptosis rate and the level of Bcl-2 in SK-NEP-1 cells.
