長鏈非編碼RNA-UBC1對膀胱癌細胞生物學功能的影響

【摘要】:目的探討長鏈非編碼RNA(lncRNA)-尿路上皮癌相關(guān)基因1(UBC1)對膀胱癌細胞增殖、凋亡和侵襲轉(zhuǎn)移的作用及其可能的機制。方法采用siRNA干擾技術(shù)沉默膀胱癌細胞系T24細胞的lncRNA-UBC1,設(shè)置陰性對照組,CCK-8檢測細胞增殖,流式細胞儀檢測細胞凋亡,Transwell檢測細胞侵襲能力,Western blot印跡法檢測與轉(zhuǎn)移相關(guān)的基質(zhì)金屬蛋白2(MMP-2)與zeste基因增強子同源2(EZH2)。結(jié)果 lncRNA-UBC1在T24細胞株中表達明顯高于人胚胎膀胱組織來源細胞;與陰性對照組和空白對照組相比,沉默UBC1后,UBC1沉默組膀胱癌細胞增殖速率慢于對照組,UBC1沉默組膀胱癌細胞凋亡細胞比例高于陰性對照組[(18.72±7.79)%vs.(13.09±5.66)%,P0.05],UBC1沉默組細胞侵襲能力受到抑制,T24細胞MMP-2與EZH2表達降低。結(jié)論沉默lncRNA-UBC1可抑制T24細胞增殖、凋亡及侵襲能力,其機制可能是通過MMP-2與EZH2途徑。
[Abstract]:Objective to investigate the effect of long chain noncoding RNA-lncRNA-UBC1 on the proliferation, apoptosis, invasion and metastasis of bladder cancer cells and its possible mechanism. Methods IncRNA-UBC1 of bladder cancer cell line T24 was silenced by siRNA interference technique, and CCK-8 was used to detect cell proliferation in negative control group. Cell apoptosis was detected by flow cytometry and the invasion ability was detected by Transwell. Western blot was used to detect the matrix metalloprotein 2 (MMP-2) associated with metastasis and zeste gene enhancer 2EZH2. Results the expression of lncRNA-UBC1 in T24 cell line was significantly higher than that in human embryonic bladder tissue derived cells, and was significantly higher than that in negative control group and blank control group. After silencing UBC1, the proliferation rate of bladder cancer cells in UBC1 silencing group was slower than that in UBC1 control group, and the percentage of apoptotic cells in UBC1 silencing group was higher than that in negative control group [18.72 鹵7.79)%vs.(13.09 鹵5.66p0.05] the cell invasion ability of UBC1 silencing group was inhibited and the expression of MMP-2 and EZH2 in UBC1 silencing group was lower than that in UBC1 silencing group. Conclusion silencing lncRNA-UBC1 can inhibit the proliferation, apoptosis and invasion of T24 cells, and its mechanism may be through MMP-2 and EZH2 pathway.
