高糖環(huán)境下纈沙坦對(duì)腎小管上皮細(xì)胞-間充質(zhì)轉(zhuǎn)化過(guò)程中骨膜蛋白、轉(zhuǎn)錄激活因子1的影響

【摘要】:目的:研究在高糖環(huán)境下纈沙坦對(duì)腎小管上皮細(xì)胞-間充質(zhì)轉(zhuǎn)化(EMT)過(guò)程中骨膜蛋白(Periostin)、轉(zhuǎn)錄激活因子1(AP-1)的影響。方法:體外高糖環(huán)境下培養(yǎng)人腎小管上皮細(xì)胞(HK-2細(xì)胞),分為空白對(duì)照組和纈沙坦高、中、低劑量組(10~(-3)、10~(-4)、10~(-5)mol/L),作用48 h;另設(shè)正常對(duì)照組。觀察各組HK-2細(xì)胞形態(tài)學(xué)變化,檢測(cè)各組HK-2細(xì)胞活力(OD值),細(xì)胞中Ⅲ型膠原含量、AP-1蛋白表達(dá)和Periostin、AP-1 mRNA表達(dá)。結(jié)果:與正常對(duì)照組比較,其余各組HK-2細(xì)胞部分細(xì)胞間隙增大并與附近的細(xì)胞脫離接觸,細(xì)胞分裂旺盛,細(xì)胞活力、Ⅲ型膠原含量、AP-1蛋白表達(dá)和Periostin、AP-1 mRNA表達(dá)均增強(qiáng)(P0.05)。與空白對(duì)照組比較,纈沙坦高、中、低劑量組細(xì)胞活力、Ⅲ型膠原含量、AP-1蛋白表達(dá)和Periostin、AP-1 mRNA表達(dá)均更低(P0.05),其中纈沙坦中劑量組作用最明顯(P0.01)。結(jié)論:體外高糖環(huán)境下可促進(jìn)HK-2細(xì)胞EMT過(guò)程;纈沙坦可通過(guò)抑制AP-1的過(guò)表達(dá)進(jìn)而下調(diào)轉(zhuǎn)分化的腎小管上皮細(xì)胞中Periostin蛋白表達(dá),從而減緩EMT過(guò)程。
[Abstract]:Objective: to study the effect of valsartan on the expression of periostein-Periostin1 (transcriptional activator 1) in the process of renal tubular epithelial cell-mesenchymal transformation (EMTT). Methods: human renal tubular epithelial cells (HK-2) were cultured in high glucose environment in vitro. Cystonia, divided into blank control group and valsartan high, In the low dose group, the cells of HK-2 were observed in morphologic changes, OD value of HK-2 cells, and the expression of AP-1 protein and PeriostinAP-1 mRNA in the cells. Results: compared with the normal control group, the expression of AP-1 protein and PeriostinAP-1 mRNA in the cells were compared with those in the normal control group, and the morphological changes of the HK-2 cells in each group were observed, and the OD value of HK-2 cells in each group was measured, and the expression of AP-1 protein and PeriostinAP-1 mRNA in the cells were compared with those in the normal control group.
