麒麟丸對少弱精子癥模型雄性大鼠生殖功能的保護作用研究

【摘要】:目的:研究麒麟丸對雷公藤多苷所致少弱精子癥模型雄性大鼠的生殖功能保護作用。方法:取28只雄性SD大鼠,隨機分成正常對照組、模型組、麒麟丸低、高劑量組,除正常對照組外,其他各組灌服雷公藤多苷40 mg/(kg·d),同時麒麟丸給藥組同日給予不同劑量麒麟丸保護,低劑量組給予麒麟丸1.62 g/(kg·d),高劑量組給予麒麟丸3.24 g/(kg·d)。連續(xù)給藥30 d后,處死大鼠,稱重大鼠睪丸,計算睪丸臟器指數(shù),檢測附睪精子濃度、活動率,檢測生殖激素水平,評估抗氧化指標,HE切片觀察大鼠睪丸組織形態(tài)學改變。結(jié)果:實驗結(jié)束后,麒麟丸低、高劑量組精子濃度分別為(14.57±3.95)×10~6/ml、(39.71±11.31)×10~6/ml,精子活力為(3.71±1.11)%、(4.29±1.80)%皆較模型組(4.71±1.25)×10~6/ml、(0.57±0.53)%有明顯提高(P0.05);與模型組FSH(1.71±0.52)mIU/ml比較,麒麟丸低、高劑量組FSH下降,分別為(1.49±0.62)mIU/ml、(1.12±0.83)mIU/ml;fT水平較模型組升高(27.27±3.63)nmol/L、(32.80±2.51)nmol/L vs(22.81±2.75)nmol/L,SHBG水平升高(94.83±11.17)nmol/L、(88.05±9.21)nmol/L vs(56.74±8.29)nmol/L,差異皆有顯著性(P0.05),但T水平無明顯變化(P0.05)。與模型組比較,麒麟丸低、高劑量組SOD水平升高(277.14±15.84)U/ml、(299.60±20.83)U/ml vs(250.04±31.06)U/ml;ROS水平下降(397.61±62.71)IU/ml、(376.84±67.14)IU/ml vs(552.20±58.07)IU/ml(P0.05)。與模型組比較,低、高劑量組HE染色顯示麒麟丸能增加少弱精子癥大鼠睪丸生精小管內(nèi)各級生精細胞層次和數(shù)量。結(jié)論:麒麟丸能明顯提高少弱精子癥大鼠精子質(zhì)量,改善生殖激素水平,減輕氧化應激損傷,改善大鼠睪丸組織形態(tài),對生殖系統(tǒng)有較好的保護作用。
[Abstract]:Objective: to study the protective effect of Kirin pill on reproductive function of male rats with oligozoospermia induced by polyglycosides of Tripterygium wilfordii. Methods: 28 male SD rats were randomly divided into normal control group, model group, low dose group and high dose group. In addition to the normal control group, the other groups were treated with Tripterygium wilfordii polyglycoside 40 mg/(kg 路dL, while the Kirin pill group was protected by different doses of Kirin pill on the same day, the low dose group was given the Kirin pill 1.62 g / kg / d, and the high dose group was given the Kirin pill 3.24 g / L / kg 路dl. The rats were killed and called testis. The index of testicular organs was calculated, the sperm concentration, activity rate, reproductive hormone level of epididymis were measured, and the antioxidation index was evaluated to observe the morphological changes of testis in rats. The sperm concentration of Qilin pill was 14.57 鹵3.95 脳 10 ~ (-6) / ml ~ (-1) 脳 10 ~ (-6) / ml, and the sperm motility was 3.71 鹵1.11 ~ 1.29 鹵1.80% respectively. Compared with model group (4.71 鹵1.25) 脳 10 ~ (6) / ml (0.57 鹵0.53)%, the sperm concentration of Kirin pill was lower than that of model group (FSH(1.71 鹵0.52mIUU / ml), and the FSH of high dose group was lower than that of model group (P < 0.05). 1.49 鹵0.62nmol / L vs(22.81 鹵2.75nmol / L vs(22.81 鹵2.75nmol / L vs(22.81 increased 94.83 鹵11.17nmol / L vs(56.74 鹵8.29nmol / L respectively, the difference was significant (P 0.05 鹵8.29nmol / L), but there was no significant change in T level. Compared with the model group, the level of SOD in the Kirin pill group was lower than that in the model group, and the SOD level in the high dose group was 277.14 鹵15.84 鹵20.83U / ml vs(250.04 鹵31.0nmol / L, which was significantly lower than that in the model group. In high dose group, he staining showed that Kirin pill could increase the level and quantity of spermatogenic cells in testicular seminiferous tubules of oligoasthenospermia rats. Conclusion: Kirin pill can obviously improve sperm quality and reproductive hormone level of oligozoospermia rats. It can reduce oxidative stress injury, improve the morphology of testis and protect the reproductive system.
