【摘要】：目的觀察miR-21在人膀胱癌5637細胞中的表達變化,探討其對細胞增殖和侵襲的影響。方法用RT-PCR技術檢測膀胱癌5637細胞和人正常膀胱上皮細胞中的miR-21表達。將培養(yǎng)好的5637細胞隨機分為轉染組、陰性對照組、空白對照組,前兩組分別轉染AS-miR-21(沉默miR-21)、無效寡核苷酸序列,空白對照組不做任何處理;分別于基因沉默后24、48、72 h采用CCK-8法測算細胞增殖指數(shù),基因沉默后24 h用Transwell侵襲實驗檢測細胞侵襲能力。結果與正常膀胱上皮細胞相比,人膀胱癌5637細胞中miR-21表達上調;沉默miR-21后,轉染組細胞增殖指數(shù)、侵襲能力低于陰性對照組、空白對照組(P均0.05)。結論 miR-21在人膀胱癌5637細胞中高表達,沉默miR-21基因后膀胱癌細胞的增殖能力和侵襲性降低。
[Abstract]:Objective to observe the expression of miR-21 in human bladder cancer cell line 5637. Methods RT-PCR technique was used to detect the expression of miR-21 in bladder cancer 5637 cells and human normal bladder epithelial cells. The machine was divided into transfection group. Negative control group, blank control group, the first two groups were transfected with AS-miR-21 (silencing miR-21), invalid oligonucleotide sequence, blank control group did not do any treatment; Cell proliferation index was measured by CCK-8 method at 24 ~ 4872 h after gene silencing. 24 h after gene silencing, Transwell invasion assay was used to detect the invasion ability of the cells. Results compared with the normal bladder epithelial cells, the expression of miR-21 was up-regulated in human bladder cancer 5637 cells. After silencing miR-21, the cell proliferation index and invasiveness of transfected group were lower than those of negative control group. Conclusion miR-21 is highly expressed in human bladder cancer 5637 cells, and the proliferation and invasiveness of bladder cancer cells are decreased after miR-21 gene silencing.
