【摘要】：目的:探討泛素連接酶(Huwe1)和小清蛋白(Parvalbumin,PV)在腦性癱瘓小鼠海馬組織中的表達(dá)及意義。方法:新生7 d昆明小鼠,隨機(jī)分為模型組和對(duì)照組,模型組按Rice的方法手術(shù)通過(guò)結(jié)扎小鼠單側(cè)頸總動(dòng)脈并缺氧來(lái)制作腦性癱瘓模型。對(duì)照組麻醉后僅切開(kāi)頸部皮膚,分離單側(cè)頸總動(dòng)脈,不結(jié)扎不剪斷,然后縫合傷口,未置于缺氧環(huán)境。各組小鼠分別在術(shù)后7 d、14 d、21 d、28 d用4%多聚甲醛灌注固定、取腦、作冰凍切片,片厚30μm,應(yīng)用免疫組化法檢測(cè)小鼠海馬組織中Huwe1和PV的表達(dá)。結(jié)果:(1)Huwe1和PV在對(duì)照組成年小鼠腦中廣泛表達(dá),在小鼠發(fā)育過(guò)程中海馬各區(qū)Huwe1和PV的表達(dá)隨著年齡的增加呈上升的趨勢(shì),至成年時(shí)趨于穩(wěn)定;(2)模型組小鼠自術(shù)后7 d起,模型組損傷側(cè)海馬CA1、CA3、DG區(qū)Huwe1的表達(dá)均高于其對(duì)側(cè)和對(duì)照組(P0.05);PV的表達(dá)雖然高于其對(duì)側(cè),卻又低于其對(duì)照組(P0.05)。結(jié)論:Huwe1在腦性癱瘓小鼠模型損傷側(cè)海馬中表達(dá)增高可能與腦損傷后其清除異常蛋白有關(guān)。PV的低表達(dá)可能與腦缺血有關(guān),其損傷側(cè)一過(guò)性的高表達(dá)提示可能與腦損傷后PV緩沖胞內(nèi)鈣超載的能力增強(qiáng)有關(guān)。
[Abstract]:Aim: to investigate the expression and significance of ubiquitin ligase Huwe1) and white protein Parvalbumin (PVV) in hippocampus of cerebral palsy mice. Methods: Kunming mice were randomly divided into model group and control group. The model group was operated on by ligating unilateral common carotid artery and hypoxia to make cerebral palsy model according to Rice's method. In the control group, the neck skin was cut open, the common carotid artery was separated, the common carotid artery was not ligated and cut, then the wound was sutured and not placed in anoxic environment. The mice in each group were fixed with 4% paraformaldehyde on the 7th day, 14th day, 21 day and 28 day after operation. The brain was taken as frozen section and the thickness of the slice was 30 渭 m. The expression of Huwe1 and PV in the hippocampus of the mice was detected by immunohistochemical method. Results the expression of Huwe1 and PV were widely expressed in the brain of adult mice in control group. The expression of Huwe1 and PV in hippocampal regions of the control group increased with the increase of age, and tended to stabilize at adulthood. In model group, the expression of Huwe1 in hippocampal CA1, CA3 and DG region was higher than that in contralateral side and control group, although it was higher than that in contralateral side, it was also lower than that in control group. Conclusion the increased expression of WHuwe1 in the injured hippocampus of cerebral palsy mice may be related to the low expression of abnormal clearance protein after brain injury, which may be related to cerebral ischemia. Transient overexpression in the injured side suggests that the increased ability of PV to buffer intracellular calcium overload may be associated with brain injury.
