螺旋藻激酶對(duì)內(nèi)皮損傷大鼠纖溶酶原和抗凝血酶Ⅲ的影響

【摘要】:目的研究螺旋藻激酶(SPK)對(duì)活體內(nèi)皮損傷大鼠血管內(nèi)皮細(xì)胞抗凝和溶栓相關(guān)因子的影響,探討SPK對(duì)心血管系統(tǒng)的保護(hù)作用。方法建立體內(nèi)大鼠內(nèi)皮損傷血栓模型。將大鼠隨機(jī)分為正常對(duì)照組、損傷模型組、SPK低、中、高劑量組。不同劑量SPK干預(yù)后麻醉動(dòng)物,Fe Cl3誘導(dǎo)大鼠頸總動(dòng)脈內(nèi)皮損傷形成血栓,從大鼠腹主動(dòng)脈取血,抗凝后用酶聯(lián)免疫吸附法(ELISA)檢測(cè)纖溶酶原(PLG)和抗凝血酶(AT)-Ⅲ的含量。結(jié)果動(dòng)物模型構(gòu)建成功后損傷模型組與正常對(duì)照組相比,大鼠血液中PLG和AT-Ⅲ的含量顯著升高(P0.01),SPK各劑量組PLG和AT-Ⅲ含量明顯升高(P0.01),在中、高劑量組最為顯著。結(jié)論 SPK在抗凝溶栓效應(yīng)上,促進(jìn)PLG和AT-Ⅲ釋放,提高內(nèi)皮細(xì)胞抗凝溶栓酶活性,從而發(fā)揮抗血栓作用。
[Abstract]:Objective to study the effect of Spirulina kinase (SPK) on the anticoagulant and thrombolytic related factors of vascular endothelial cells in rats with living endothelial injury and to explore the protective effect of SPK on cardiovascular system. Methods the rat model of endothelium injury thrombus was established in vivo. The rats were randomly divided into normal control group, damage model group, low SPK, high dose group and different dose of SPK. After the intervention of the anesthetized animals, Fe Cl3 induced the endothelial injury of the common carotid artery to form a thrombus, the blood was taken from the abdominal aorta of the rat, and the content of the plasminogen (PLG) and the antithrombin (AT) - III were detected by the enzyme linked immunosorbent assay (ELISA) after anticoagulation. The results of the animal model group were compared with the normal control group, and the PLG and AT- in the rat blood were compared with the normal control group. The content of III was significantly increased (P0.01), and the content of PLG and AT- III in each dose group of SPK increased significantly (P0.01). In the middle, the high dose group was the most significant. Conclusion SPK in anticoagulant thrombolytic effect, promote the release of PLG and AT- III, improve the activity of anticoagulant thrombolytic enzyme in endothelial cells, and thus play an anti thrombolytic effect.
