丙泊酚對腦缺血再灌注大鼠腦組織Homer-1a表達的影響及其機制

【摘要】:目的觀察丙泊酚對腦缺血再灌注大鼠腦組織Homer-1a表達的影響,并探討其機制。方法雄性SD大鼠120只,隨機分為假手術組、造模組、丙泊酚組,每組40只。模型組、丙泊酚組線栓法建立腦缺血再灌注模型,假手術組僅行血管分離處理不插入線栓;造模后30 min,丙泊酚組腹腔注射丙泊酚10 mg/100 g,假手術組和對照組均腹腔注射等體積生理鹽水。給藥處理3 h、24 h、72 h、7 d分別進行神經功能評分,采用Real-time PCR法檢測腦組織Homer-1a mRNA,應用酶聯(lián)免疫吸附法檢測血清S-100β。結果與假手術組比較,模型組和丙泊酚組4個時間點神經功能評分、血清S-100β水平均增加(P均0.05),以模型組增高更顯著(P均0.05)。與假手術組相比,模型組在3、24 h時間點腦組織Homer-1a mRNA降低(P均0.05);丙泊酚組4個時間點腦組織Homer-1a mRNA均較其他兩組升高(P均0.05)。在所有大鼠樣本中,腦組織Homer-1a mRNA表達與血清S-100β呈負相關(r=-0.939,P0.01)。結論丙泊酚麻醉可通過誘導腦組織Homer-1a mRNA表達發(fā)揮對腦缺血再灌注大鼠的腦保護作用,該作用可能與其降低血清S-100β水平有關。
[Abstract]:Objective to observe the effect of propofol on the expression of Homer-1a in cerebral ischemia reperfusion rats and to explore its mechanism. Methods 120 male SD rats were randomly divided into sham-operated group, model group and propofol group with 40 rats in each group. Model group, propofol group was used to establish cerebral ischemia-reperfusion model, sham-operation group was only treated with vascular separation and did not insert thread embolus. After 30 minutes, propofol was injected intraperitoneally with propofol for 10 mg/100 g, and normal saline was injected intraperitoneally in sham operation group and control group. The nerve function scores were evaluated by Real-time PCR method and serum S-100 尾 was detected by enzyme-linked immunosorbent assay (Elisa). Results compared with the sham operation group, the nerve function scores and serum S-100 尾 levels in the model group and propofol group were increased at 4 time points (P < 0.05), especially in the model group (P = 0.05). Compared with the sham operation group, the Homer-1a mRNA of brain tissue in the model group decreased by 0.05 at 3h at 24 h, and the Homer-1a mRNA in the brain tissue in the propofol group at four time points was higher than that in the other two groups (P < 0.05). There was a negative correlation between the expression of Homer-1a mRNA and serum S-100 尾 in all rat samples. Conclusion propofol anesthesia can induce the expression of Homer-1a mRNA in brain tissue and play a protective role in cerebral ischemia-reperfusion rats, which may be related to the decrease of serum S-100 尾 level.
