三種修復(fù)粘結(jié)材料對(duì)L929細(xì)胞毒性的研究

【摘要】:[目的]通過(guò)CCK-8法和流式細(xì)胞術(shù)的檢測(cè),觀察了解玻璃離子水門(mén)汀、聚羧酸鋅水門(mén)汀、3M自粘結(jié)樹(shù)脂粘固劑三種粘結(jié)材料對(duì)小鼠成纖維細(xì)胞L929的細(xì)胞增殖率和細(xì)胞凋亡率的影響及差異,為臨床應(yīng)用提供實(shí)驗(yàn)依據(jù)。[方法]用CCK-8法檢測(cè)三種粘結(jié)材料浸提3d、7d、14d三個(gè)時(shí)間段的細(xì)胞增殖率(relative growth rate,RGR)和流式細(xì)胞術(shù)檢測(cè)三種粘結(jié)材料浸提3d、7d兩個(gè)時(shí)間段的細(xì)胞凋亡率,觀察三種粘結(jié)材料對(duì)小鼠成纖維細(xì)胞L929的影響和差異。[結(jié)果]1.玻璃離子水門(mén)汀的 RGR 分別為 86.84%(3d)、82.85%(7d)、84.27%(14d),細(xì)胞毒性均為1級(jí),表現(xiàn)為輕微細(xì)胞毒性。不同浸提時(shí)間組之間無(wú)明顯差異(P0.05)。2.聚羧酸鋅水門(mén)汀的 RGR 分別為 79.26%(3d)、77.93%(7d)、72.47%(14d),3d和7d細(xì)胞毒性為1級(jí),14d細(xì)胞毒性為2級(jí),表現(xiàn)為輕微細(xì)胞毒性。不同浸提時(shí)間組之間無(wú)明顯差異(P0.05)。3.3M自粘結(jié)樹(shù)脂粘固劑的RGR分別為114.34%(3d)、116.57%(7d)、114.84%(14d),細(xì)胞毒性為0級(jí),未表現(xiàn)出細(xì)胞毒性。不同浸提時(shí)間組之間無(wú)明顯差異(P0.05)。[結(jié)論]1.聚羧酸鋅水門(mén)汀3d,7d兩個(gè)時(shí)間組的細(xì)胞毒性為1級(jí),表現(xiàn)輕微細(xì)胞毒性;14d時(shí)間組的細(xì)胞毒性為2級(jí),表現(xiàn)為輕度細(xì)胞毒性;2.玻璃離子水門(mén)汀組三個(gè)時(shí)間組的細(xì)胞毒性均為1級(jí),表現(xiàn)出輕微的細(xì)胞毒性;3.3M自粘結(jié)樹(shù)脂粘固劑的細(xì)胞毒性則為0級(jí),無(wú)細(xì)胞毒性;4.三種粘結(jié)材料均滿足口腔材料臨床應(yīng)用的生物相容性的要求。
[Abstract]:[objective] to observe and understand glass ionomer cement by CCK-8 method and flow cytometry. Effects and differences of three kinds of adhesive materials on cell proliferation and apoptosis of mouse fibroblasts L929, which provides experimental basis for clinical application. [methods] the cell proliferation rate and relative growth rateRGRs of three kinds of adhesive materials were measured by CCK-8 and flow cytometry respectively. The apoptotic rates of three kinds of adhesive materials were detected at 3 days and 7 days after extraction. The effects and differences of three kinds of adhesive materials on mouse fibroblasts L 929 were observed. [result] 1. The RGR of vitreous ionomer cement was 86.84 d ~ 82.85 and 84.27 ~ 14 d ~ (-1) respectively. The cytotoxicity of glass ionomer was grade 1, showing slight cytotoxicity. There was no significant difference between different extraction time groups. The RGR of zinc polycarboxylate cement was 79.26 days and 77.93 days, respectively. The cytotoxicity of zinc polycarboxylate was 72.47 days and 14days respectively. The cytotoxicity was grade 1, grade 14, and grade 2, showing slight cytotoxicity. There was no significant difference between different extraction time groups. The RGR of self-adhesive resin binder was 114.343d ~ 116.577d and 114.84d ~ 14d, respectively. The cytotoxicity was 0 grade, no cytotoxicity was found. There was no significant difference between different extraction time groups (P 0.05). [conclusion] 1. The cytotoxicity of zinc polycarboxylate cement for 3 days and 14 days was grade 1 and grade 2, respectively. The cytotoxicity of glass-ionomer cement group was grade 1, and the cytotoxicity of glass-ionomer cement group was grade 0 and no cytotoxicity. All three kinds of adhesive materials meet the requirements of biocompatibility for clinical application of oral materials.
