【摘要】：目的探討Kv1.3在人牙周膜成纖維細(xì)胞(h PDLFs)增殖和分化成骨能力中的作用。方法通過(guò)體外分離并培養(yǎng)健康人和牙周炎患者牙周膜成纖維細(xì)胞,應(yīng)用細(xì)胞免疫熒光法、Q-PCR和Western blot檢測(cè)Kv1.3在牙周炎h PDLFs中表達(dá)變化;并通過(guò)si RNA沉默抑制Kv1.3表達(dá),使用MTT和流式細(xì)胞儀觀察對(duì)h PDLFs的增殖和分化成骨能力的作用。結(jié)果細(xì)胞免疫熒光結(jié)果顯示與健康組比較,牙周炎組h PDLFs中Kv1.3顯著低表達(dá)(P0.05);Western blot結(jié)果顯示Kv1.3-si RNA能夠明顯抑制細(xì)胞周期蛋白D1(Cyclin D1)和C-myc蛋白表達(dá)(P0.05);流式細(xì)胞儀檢測(cè)顯示Kv1.3-si RNA能夠顯著抑制細(xì)胞G2/M期;同時(shí),Western blot結(jié)果顯示Kv1.3-si RNA明顯降低堿性磷脂酶(ALP)和骨鈣素(OCN)蛋白表達(dá)。結(jié)論抑制Kv1.3表達(dá)能夠明顯抑制h PDLFs的增殖和分化成骨能力。
[Abstract]:Objective to investigate the role of Kv1.3 in the proliferation and differentiation of human periodontal ligament fibroblasts (PDLFs). Methods periodontal ligament fibroblasts were isolated and cultured from healthy subjects and patients with periodontitis in vitro. The expression of Kv1.3 in PDLFs of periodontitis was detected by Q-PCR and Western blot, and the expression of Kv1.3 was inhibited by si RNA silencing. The effects of MTT and flow cytometry on the proliferation and differentiation of h PDLFs were observed. Results the results of cellular immunofluorescence showed that compared with the healthy group, In the periodontitis group, the expression of Kv1.3 in PDLFs was significantly lower than that in the control group. The results of Western blot showed that Kv1.3-si RNA could significantly inhibit the expression of cyclin D1(Cyclin D1) and C-myc protein, and the flow cytometry showed that Kv1.3-si RNA could significantly inhibit the G 2 / M phase of the cells. At the same time, the results of Western blot showed that Kv1.3-si RNA significantly decreased the expression of alkaline phospholipase (ALP) and osteocalcin (OCN) protein. Conclusion inhibition of Kv1.3 expression can significantly inhibit the proliferation and differentiation of h PDLFs.
