【摘要】：目的:對人牙周膜干細胞進行篩選純化,骨向誘導后觀察其成骨能力,并對成骨相分化過程中相關基因的表達變化進行檢測。方法:用組織塊聯(lián)合磁珠分選法分離純化人牙周膜干細胞,初步探討其成骨性能,堿性磷酸酶(ALP)和茜素紅染色觀察其成骨情況,實時定量PCR檢測其相關成骨基因的表達變化。利用SPSS12.0軟件包對數(shù)據(jù)進行統(tǒng)計學分析。結(jié)果:組織塊聯(lián)合磁珠分選法能成功培養(yǎng)出高純度的人牙周膜干細胞。體外培養(yǎng)中,PDLSCs具備成脂和成骨的雙相分化能力。在成骨誘導過程中,FOXO1和RUNX2的表達先升高再降低,ALP和OCN的基因表達水平持續(xù)增高。結(jié)論:成骨相關基因ALP、RUNX2、FOXO1和OCN均參與其向成骨樣組織分化的過程,其表達變化具有時間規(guī)律性,與成骨過程類似。
[Abstract]:Objective: in human periodontal ligament stem cells were purified to observe the bone after induction of osteogenesis, and osteogenic differentiation phase expression of relevant genes in the process of testing. Methods: isolation and purification of human periodontal ligament stem cells with tissue combined with Macs, to explore its osteogenic properties. Alkaline phosphatase (ALP) and alizarin red staining to observe the osteogenesis, real-time quantitative PCR to detect the expression of genes related to bone. Packet data were analyzed by SPSS12.0 software. Results: the tissue block combined with Macs can successfully produce high purity human periodontal ligament stem cells. In vitro. PDLSCs with the adipogenic and osteogenic differentiation ability in dual phase. During osteogenic induction, the expression of FOXO1 and RUNX2 increased first and then decreased, ALP and OCN gene expression level continues to increase. Conclusion: the osteogenic related genes ALP, RUNX2, FOXO1 and OCN are involved in In the process of osteogenic tissue differentiation, its expression changes have time regularity, which is similar to the process of osteogenesis.
