  血管性認(rèn)知功能障礙(vascular cognitive impairment,VCI)是由各種腦血管疾病引起的以學(xué)習(xí)、記憶功能障礙為主要癥狀的獲得性智能損害綜合征,其病程發(fā)展的晚期階段被稱作血管性癡呆(vascular dementia,VD)。目前,VD已經(jīng)成為世界范圍內(nèi)繼阿爾茨海默病(Alzheimer’s disease,AD)之后的第二大癡呆類型。但是,迄今為止,VD的發(fā)病機(jī)制尚不完全明確,因而缺乏有效的治療藥物。研究表明,慢性腦低灌注(chronic cerebral hypoperfusion,CCH)損傷在VD的發(fā)病過(guò)程中發(fā)揮著重要作用,因此探討慢性腦低灌注損傷導(dǎo)致VD的發(fā)病機(jī)制,并尋求有效的癡呆防治藥物成為現(xiàn)代醫(yī)學(xué)的研究熱點(diǎn)之一。已有的研究表明,神經(jīng)元的自噬與凋亡在慢性腦低灌注損傷導(dǎo)致VD的發(fā)病機(jī)制中占有重要地位,因此調(diào)控神經(jīng)元的自噬與凋亡對(duì)于減緩神經(jīng)元損傷至關(guān)重要。磷脂酰肌醇-3-激酶/蛋白激酶B(phosphoinositide-3-kinase/protein kinase B,PI3K/Akt)信號(hào)通路是一條經(jīng)典的抗凋亡、促存活通路,該通路的激活在缺血缺氧性神...
